The nuclear matrix-intermediate filament system of human neuroblastoma SK-N-SH cells before and after retinoic acid (RA) treatment was selectively extracted and the distribution of prohibitin (PHB) in the nuclear matrix, as well as its colocalization with related genes, was observed. Results of two-dimensional gel electrophoresis (2-DE), mass spectrometry (MS) identification, and protein immunoblotting all confirm that PHB was present in the components of SK-N-SH nuclear matrix proteins and was down-regulated after RA treatment. Immunofluorescence microscopy observations show that PHB was localized in the nuclear matrix and its distribution was altered due to RA treatment. Laser confocal microscopy results reveal that PHB colocalized with the expression products of c-myc, c-fos, p53, and Rb, but the colocalization region was altered after RA treatment. Our results prove that PHB is a nuclear matrix protein and is localized in nuclear matrix fibers. The distribution of PHB in SK-N-SH cells and its colocalization with related proto-oncogenes and tumor suppressor genes suggest that PHB plays pivotal roles in the differentiation of SK-N-SH cells and deserves further study.

Aortic valve stenosis is a progressive and increasingly prevalent disease in older adults, with no approved pharmacologic therapies to prevent or slow its progression. Although genetic risk factors have been identified, the contribution of epigenetic regulation remains poorly understood. Here, we demonstrated that histone deacetylase 3 (HDAC3) maintains aortic valve structure by suppressing mitochondrial biogenesis and preserving extracellular matrix integrity in valvular interstitial fibroblasts.

Thyroid cancer, a prevalent endocrine malignancy, is influenced by its tumor microenvironment (TME), with cancer-associated fibroblasts (CAFs) playing a pivotal role in disease progression. Molecularly, CAFs orchestrate a pro-tumorigenic niche via cytokine secretion and extracellular matrix (ECM) stiffening, underscoring their targetability. Therapeutic strategies, including small molecule inhibitor-based therapies, immune-based therapies, nanoparticle-based approaches, and combination regimens, have been evaluated for their efficacy in disrupting CAF functionality.

In this study, a one-pot hydrothermal synthesis method was used to synthesize a novel gold-yttrium trimesic acid metal-organic framework (Au-Y-TMA MOF), demonstrating significant improvements over conventional single-metal MOFs, that is, yttrium trimesic acid (Y-TMA), in both supercapacitor applications and electrochemical antibiotic detection. The X-ray diffraction patterns of Au-Y-TMA confirmed the presence and impact of Au in the Y-TMA matrix, while field emission scanning electron microscopy (FE-SEM) images revealed a heterogeneous combination of gold nanoparticles (AuNPs) and Y-TMA, suggesting a nonuniform distribution and possible interaction. The developed half-cell supercapacitor exhibited a remarkable capacitance value of 1836 F/g at a current density of 5 A/g by galvanostatic charging-discharging (GCD) measurement.

This paper develops an inferential framework for matrix completion when missing is not at random and without the requirement of strong signals. Our development is based on the observation that if the number of missing entries is small enough compared to the panel size, then they can be estimated well even when missing is not at random. Taking advantage of this fact, we divide the missing entries into smaller groups and estimate each group via nuclear norm regularization.

Introduction: Age-related macular degeneration (AMD) is a leading cause of vision loss in older adults, with limited effective treatments available. This study aimed to investigate the pharmacological effects of dihydromyricetin (DHM) on AMD and to identify its putative pharmacological targets through network analysis and molecular docking approaches.
Methods: experiments established an AMD model using sodium iodate (SI)-induced ARPE-19 cells, with CCK-8 assays determining 15 mM SI as the optimal modeling concentration and 100 μM DHM as the optimal treatment concentration.
