Article Abstract

Biaryl ethers were recently reported as potent NNRTIs. Herein, we disclose a detailed effort to modify the previously reported compound 1. We have designed and synthesized a series of novel pyrazole derivatives as a surrogate for pyrazolopyridine motif that were potent inhibitors of HIV-1 RT with nanomolar intrinsic activity on the WT and key mutant enzymes and potent antiviral activity in infected cells.

A visible-light-promoted copper-catalyzed C-O coupling reaction of alcohols with aryl bromides using eosin Y as the photosensitizer has been developed, in which energy transfer (EnT) from eosin Y to the copper complex of the oxalamide ligand generates an excited-state copper complex that facilitates oxidative addition of aryl bromides. This mild method enables the efficient synthesis of alkyl aryl ethers and biaryl ethers with a broad substrate scope and excellent tolerance of sensitive functional groups at room temperature.

We report an ether-directed enantioselective C(sp)-H borylation catalyzed by a chiral bidentate boryl ligand (CBL)/iridium system for constructing axially chiral biaryls. This method delivered diverse chiral biaryls with good to high enantioselectivities, accommodating varied electronic and steric substituents on the aryl rings. Gram-scale synthesis and downstream transformations of the C-B bond underscored its practicality.

Highly encumbered 2,2',6-tri- and 2,2',6,6'-tetra-substituted biaryls are readily prepared from aryl -iodobenzyl ethers through mediated cathodic reduction under flow. The reaction proceeds the stepwise transfer of two electrons: the first to induce loss of iodide and a radical cyclisation, and the second to induce a polar fragmentation.

Despite recent advances in the inhibition of EGFR (epidermal growth factor receptor), there remains a clinical need for new EGFR Exon20 insertion (Ex20Ins) inhibitors that spare EGFR WT. Herein, we report the discovery and optimization of two chemical series leading to ether and biaryl as potent, selective, and brain-penetrant inhibitors of Ex20Ins mutants. Building on our earlier discovery of alkyne which allowed access to CNS property space for an Ex20Ins inhibitor, we utilized structure-based design to move to lower lipophilicity and lower CL compounds while maintaining a WT selectivity margin.

ConspectusChiral organosilicon compounds bearing a Si-stereogenic center have attracted increasing attention in various scientific communities and appear to be a topic of high current relevance in modern organic chemistry, given their versatile utility as chiral building blocks, chiral reagents, chiral auxiliaries, and chiral catalysts. Historically, access to these non-natural Si-stereogenic silanes mainly relies on resolution, whereas their asymmetric synthetic methods dramatically lagged compared to their carbon counterparts. Over the past two decades, transition-metal-catalyzed desymmetrization of prochiral organosilanes has emerged as an effective tool for the synthesis of enantioenriched Si-stereogenic silanes.
