Neuron-glia interaction is involved in physiological function of neurons, however, recent evidences have suggested glial cells as participants in neurotoxic and neurotrophic mechanisms of neurodegenerative/neuroregenerative processes. Laser microdissection offers a unique opportunity to study molecular regulation in specific immunolabeled cell types. However, an adequate protocol to allow morphological and molecular analysis of rodent spinal cord astrocyte, microglia and motoneurons remains a big challenge. In this paper we present a quick method to immunolabel those cells in flash frozen sections to be used in molecular biology analyses after laser microdissection and pressure catapulting.

Immunoelectron Microscopy (IEM) is a technique that combines specific immunolabeling with high-resolution electron microscopic imaging to achieve precise spatial localization of biomolecules at the subcellular scale (< 10 nm) by using high-electron-density markers such as colloidal gold and quantum dots. As a core tool for analyzing the distribution of proteins, organelle interactions, and localization of disease pathology markers, it has irreplaceable value, especially in synapse research, pathogen-host interaction mechanism, and tumor microenvironment analysis. Background: Tissue clearing techniques combined with light-sheet fluorescence microscopy (LSFM) enable high-resolution 3D imaging of biological structures without physical sectioning. While widely used in neuroscience to determine brain architecture and connectomics, their application for spinal cord mapping remains more limited, posing challenges for studying demyelinating diseases like multiple sclerosis. Myelin visualization in cleared tissues is particularly difficult due to the lipid-removal nature of most clearing protocols, and alternative immunolabeling approaches failed to reach satisfying results.

Background And Purpose: Cisplatin is a widely used chemotherapy drug for the treatment of solid tumours, but its clinical benefit is often limited by ototoxicity, leading to irreversible sensorineural hearing loss. However, there is a lack of effective strategies to prevent hearing loss caused by cisplatin in adults, while sodium thiosulfate is approved by the Food and Drug Administration in the United States for only use at the pediatric level. Sarsasapogenin, a natural compound of the , has antioxidant and neuroprotective properties, which suggest that it may attenuate the ototoxicity induced by cisplatin.
