Aberrant activation of fibroblast growth factor receptors (FGFRs) plays a critical role in tumorigenesis across multiple cancer types, driving the development of various FGFR inhibitors. Despite clinical advances, therapeutic efficacy remains limited by the emergence of drug resistance, primarily mediated by gatekeeper mutations in FGFRs. To overcome this challenge, we designed and synthesized a novel series of 7-(1-methyl-1-indole-3-yl)-5-pyrrolo[2,3-]pyrazine derivatives as covalent pan-FGFR inhibitors targeting both wild-type and gatekeeper mutants.

Material And Methods: We included patients who underwent surgical intervention for tethered cord in our clinic between 2010 and 2020 and were subsequently diagnosed with retethering during follow-up. Only those with available postoperative clinical follow-up data were included.

Neurogenic bladder and lower urinary tract (LUT) dysfunctions encompass a wide variety of urinary disorders resulting from nervous system impairments. Unfortunately, conventional treatments are still limited and can have significant complication rates, especially when stent implantations or other surgical procedures are involved. Therefore, there is a critical need to develop novel therapeutic strategies and pharmacological approaches to address these challenging urological conditions.

Background: Due to the complex structure and variable microenvironment in the progression of bladder cancer, the efficacy of traditional treatment methods such as surgery and chemotherapy is limited. Tumor residual, recurrence and metastasis are still difficult to treat. The integration of diagnosis and treatment based on nanoparticles can offer the potential for precise tumor localization and real-time therapeutic monitoring.
