Xylella fastidiosa (Xf) is a xylem-limited bacterial pathogen that causes leaf scorch symptoms in numerous plant species in urban, agricultural, and natural ecosystems worldwide. The exact mechanism of hydraulic disruption and systemic colonization of xylem by Xf remains elusive across all host plants. To understand both processes better, the functional and structural characteristics of xylem in different organs of both healthy and Xf-infected trees of several Quercus species were studied. Hydraulic conductivity (K(s)) in Xf-infected petioles of Q. palustris and Q. rubra decreased significantly compared with healthy trees as the season progressed and plummeted to zero with the onset of scorch symptoms. Prior to the onset of symptoms, embolism was as much as 3.7 times higher in Xf-infected petioles compared with healthy controls and preceded significant reductions in K(s). Embolism likely resulted from pit membrane degradation during colonization of new petiole xylem and triggered the process that eventually led to vessel occlusion. Pit membrane porosity was studied using the following four methods to determine if a pathway exists in the xylem network of woody stems that allows for passive Xf migration: (i) calculations based on vulnerability to cavitation data, (ii) scanning electron micrographs, (iii) microsphere injections, and (iv) air seeding thresholds on individual vessels. All four methods consistently demonstrated that large pit membrane pores (i.e. greater than the diameter of individual Xf) occur frequently throughout the secondary stem xylem in several Quercus species. These large pores probably facilitate systemic colonization of the secondary xylem network and contribute to the high susceptibility to bacterial leaf scorch exhibited among these species.

Purpose: To reassess the anatomic basis of optic disc pit maculopathy (OPM) using swept-source optical coherence tomography (SS-OCT) and to characterize the broader structural abnormalities comprising the optic pit complex.
Methods: Sixteen patients with OPM were imaged using a high-resolution SS-OCT system (DREAM OCT). Cross-sectional and volume-rendered scans were analyzed for lamina cribrosa defects, intraneural cavitations, and pathways for fluid entry into or beneath the retina.

Optic disc pit maculopathy is a serious complication of congenital optic disc pits, often resulting in significant vision loss due to intraretinal and subretinal fluid accumulation. This case report describes a 35-year-old woman with progressive visual decline in her right eye. Examination revealed optic disc pit maculopathy with marked central foveal thickness (634 μm) and a best-corrected visual acuity of 20/100.

Near-infrared photoimmunotherapy (NIR-PIT) is a highly selective cancer therapy based on a targeted monoclonal antibody conjugated to phthalocyanine-based photoabsorber, IRDye700DX (IR700). Following injection, near-infrared light activates the conjugate causing direct cytotoxicity and immunogenic cell death. The high selectivity of NIR-PIT is traditionally based on the binding of a monoclonal antibody to its target thus bringing the photoabsorber, IR700, in close proximity to the cell membrane at the time of light exposure.

Fusobacterium nucleatum is overrepresented in the colon microbiome of colorectal cancer patients and has been associated with tumor growth enhancement and metastasis. A pivotal pathogenic factor, the autotransporter adhesin Fap2, facilitates association to cancer and immune cells via the receptors Gal-GalNAc and TIGIT, respectively, leading to deactivation of immune cells. Mechanistic details of the Fap2/TIGIT interaction remain elusive as no structural data are available.
