With the approval of the antibody-drug conjugate enfortumab vedotin (EV), NECTIN4 has emerged as a bona fide therapeutic target in urothelial carcinoma (UC). Here, we report the development of a NECTIN4-directed chimeric antigen receptor (CAR) T cell, which exhibits reactivity across cells expressing a range of endogenous NECTIN4, with enhanced activity in high expressors. We demonstrate that the PPARγ pathway, critical for luminal differentiation, transcriptionally controls NECTIN4, and that the PPARγ agonist rosiglitazone primes and augments NECTIN4 expression, thereby increasing sensitivity to NECTIN4-CAR T cell-mediated killing.

Histone methylation (HMT), the enzymatic addition of methyl groups to specific histone residues by histone methyltransferases, constitutes a key regulatory mechanism in gene expression and post-translational modulation. Although studies have explored HMT's role in oncogenesis and other organ-specific disorders, HMT is now implicated in the pathogenesis of kidney diseases. A broad spectrum of experimental models, including both and systems, has demonstrated the involvement of HMT alterations in diverse renal pathologies such as acute kidney injury, renal fibrosis, diabetic nephropathy, lupus nephritis, polycystic kidney disease, kidney stones, renal cell carcinoma, and immunoglobulin A nephropathy.

Sheep milk has gained increasing attention for its compositional similarity to human milk and its abundance of bioactive compounds with nutritional and therapeutic potential. It is rich in proteins, essential fatty acids, vitamins, minerals, immunoglobulins, and hormones, as well as peptides and oligosaccharides with antiviral, antibacterial, anti-inflammatory, and immune-modulatory effects. Despite these benefits, the literature remains fragmented, with limited integration of data on the mechanisms by which these components influence health outcomes, and few comprehensive comparisons with other mammalian milks.

This study aimed to assess the impact of yeast beta-1,3/1,6-glucans (BG) on apparent digestibility coefficients (ADC) of nutrients, intestinal fermentative metabolites, fecal microbiota profile, and immune and antioxidant variables in puppies before and after surgical challenge. Two treatments were evaluated: control, without, and test, with oral supplementation of 65 mg/kg body weight/day of purified BG from Saccharomyces cerevisiae for 120 days.
