Article Abstract

Aim: To investigate the action of genistein (GST), a broad spectrum tyrosine kinase inhibitor, on voltage-gated potassium channels in guinea pig proximal colon smooth muscle cells.

Methods: Smooth muscle cells in guinea pig proximal colon were enzymatically isolated. Nystatin-perforated whole cell patch clamp technique was used to record potassium currents including fast transient outward current (IKto) and delayed rectifier current (IKdr), two of which were isolated pharmacologically with 10 mmol/L tetraethylammonium or 5 mmol/L 4-aminopyridine. Contamination of calcium-dependent potassium currents was minimized with no calcium and 0.2 mmol/L CdCl2 in an external solution.

Results: GST (10-100 micromol/L) reversibly and dose-dependently reduced the peak amplitude of IKto with an IC50 value of 22.0+/-6.9 micromol/L. To a lesser extent, IKdr was also inhibited in both peak current and sustained current. GST could not totally block the outward potassium current as a fraction of the outward potassium current, which was insensitive to GST. GST had no effect on the steady-state activation (n=6) and inactivation kinetics (n=6) of IKto. Sodium orthovanadate (1 mmol/L), a potent inhibitor of tyrosine phosphatase, significantly inhibited GST-induced inhibition (P<0.05).

Conclusion: GST can dose-dependently and reversibly block voltage-gated potassium channels in guinea pig proximal colon smooth muscle cells.

Background: Fetal growth restriction (FGR) causes an adaptive redistribution of the cardiac output towards sustained cerebral vasodilation. However, the consequences of FGR and cerebral vasodilatation due to fetal hypoxia on the blood-brain barrier (BBB) are still poorly studied. This study assesses BBB permeability in the neonatal cortex of pups gestated under intrauterine hypobaric hypoxia.

The high prevalence and drug resistance of superficial fungal infections (SFIs) pose a serious global public health challenge, urgently necessitating the development of novel antifungal drugs with unique structures and novel targets. Evodiamine derivatives are promising antifungal drug leads, yet they suffer from suboptimal activity and unclear action mechanisms. In this study, a series of N(14)-phenyl evodiamine derivatives were designed, synthesized and tested for their biological activities.

Rodent adapted marburg viruses are lethal in ferrets.

Special Pathogens Program, National Microbiology Laboratory Branch, Public Health Agency of Canada, Winnipeg, MB, Canada.

Ferrets are highly susceptible to infection with several orthoebolaviruses, including Ebola virus (EBOV), yet they are refractory to infection with the orthomarburgviruses, Marburg virus (MARV) and Ravn virus. This study sought to investigate the pathogenicity of rodent-adapted MARV in ferrets. Challenge with guinea pig-adapted (GPA)-MARV resulted in uniform lethality among ferrets, whereas challenge with mouse-adapted (MA)-MARV resulted in partial lethality.

The low dose dexamethasone stimulation test (LDDST) is routinely used in canine medicine but in the few cases in guinea pigs diagnosed with hypercortisolism, the adrenocorticotropic hormone (ACTH) stimulation test was used. The objective of the authors was to conduct a pilot study and find out if the standard test used in dogs can be used in this species.A 4-year-old intact female hairless guinea pig showed bilaterally enlarged adrenal glands and high cortisol levels.

Purpose: To evaluate the efficacy and underlying mechanism of advanced optimal pulse technology intense pulsed light (AOPT) in low-energy triple-pulse long-width mode (AOPT-LTL) for melasma treatment.

Methods: An in vivo guinea pig model of melasma was established through progesterone injection and ultraviolet B radiation. Three sessions of AOPT-LTL treatment were performed weekly.
