Article Abstract

In this study, three epigallocatechin gallate glycosides were synthesized by the acceptor reaction of a glucansucrase produced by Leuconostoc mesenteroides B-1299CB with epigallocatechin gallate (EGCG) and sucrose. Each of these glycosides was then purified, and the structures were assigned as follows: epigallocatechin gallate 7-O-alpha-D-glucopyranoside (EGCG-G1); epigallocatechin gallate 4'-O-alpha-D-glucopyranoside (EGCG-G1'); and epigallocatechin gallate 7,4'-O-alpha-D-glucopyranoside (EGCG-G2). One of these compounds (EGCG-G1) was a novel compound. The EGCG glycosides exhibited similar or slower antioxidant effects, depending on their structures (EGCG > or = EGCG-G1 > EGCG-G1' > EGCG-G2), and also manifested a higher degree of browning resistance than was previously noted in EGCG. Also, EGCG-G1, EGCG-G1', and EGCG-G2 were 49, 55, and 114 times as water soluble, respectively, as EGCG.

The discovery of flavonoid amination metabolism has raised research interest in this new biotransformation mechanism. studies have revealed that flavonoids with pyrogallol structures readily react with N-nucleophilic ammonia, where amination occurs at intermediate -OH position, such as the B ring C4'-OH of epigallocatechin gallate and myricetin, A ring C6-OH of baicalein. Flavonoids are also covalently bound with amino acid residue by Schiff base, Michael addition or Strecker degradation, which are further rearranged to generate -NH substituted products.

Dentin caries is a multifactorial pathological process characterized by bacterial colonization and biofilm formation that result in concurrent acid-mediated demineralization and matrix metalloproteinase (MMP)-mediated degradation of the collagenous matrix. While remineralization therapies offer minimal invasiveness, their long-term efficacy is compromised by ongoing collagen degradation and persistent bacterial acid production that counteract remineralization efforts. To address these limitations, we designed PAMAM-G4@EG (PGE) nanoparticles (NPs) using polyamide amine (PAMAM) dendrimers as mineral deposition templates, with antimicrobial peptide G(IIKK)I-NH (G4) grafted onto the external surface groups and epigallocatechin gallate (EG) encapsulated within the internal cavities to provide biofilm disintegration and collagen protection for comprehensive dentin caries intervention.
