Objective: To investigate the role of Ras antisense oligoribonucleotide (ASODN) in multidrug resistance (MDR) of pancreatic carcinoma Pc-2 cells.
Methods: Ras and P-gp expression was suppressed by Ras ASODN. Sensitivity of Pc-2 cells to chemotherapy was determined by the MTT assay. MDR-1 mRNA level was detected by fluorogenic probe quantitative reverse transcription polymerase chain (RT-PCR) method. Flow cytometry (FCM) was used to detect the accumulative concentration of adriamycin (ADR) in the cells.
Results: Ras ASODN significantly inhibited the Ras and P-gp expression (P < 0.05), increased the sensitivity of Pc-2 cells to chemotherapeutic agents (P < 0.05), decreased MDR-1 gene level in Pc-2 cells (P < 0.05), and increased the intracellular intake of ADR in Pc-2 cells (P < 0.05).
Conclusion: Ras ASODN may enhance the sensitivity of multidrug-resistant pancreatic cancer Pc-2 cells to chemotherapeutic agents by regulating MDR-1 gene level.

Luminescent solar concentrators (LSCs) offer a sustainable approach to power generation using fluorescent glasses, yet their green industrialization is impeded by the limited production scale and non-recyclability of embedded nanocrystals. Here, we introduce a lead-free perovskite derivative ETPSbCl (ETP = (CH)PCH) with a reversible transition between powder and glass states. Through molecular dynamics and density functional theory, we elucidate the possible structural distortions of [SbCl] pyramids and their impact on luminescence.

The endoplasmic reticulum (ER) is a highly dynamic organelle that undergoes significant morphological alterations in response to cellular stress. While conventional transmission electron microscopy (TEM) has provided valuable insights into these changes, such as the formation of crystalloid-ER and ER whorls, obtaining comprehensive three-dimensional (3D) information on these large structures within their cellular context has remained a challenge. To overcome these limitations, this study introduces an innovative application of dual-axis scanning transmission electron microscopy (STEM) tomography to investigate ER morphology under stress conditions in human embryonic kidney (HEK) cells overexpressing the cation channel polycystin-2 (PC-2).

The preparation of new photosensitizers and studies on photodynamic therapy (PDT) have provided promising results and realistic expectations for an efficient medical treatment. Phthalocyanines (Pcs) have become extremely attractive for this purpose owing to their molecular versatility and superior physicochemical properties. Motivated by these facts, in this study, non-peripherally substituted zinc(II) Pc (2) was prepared by cyclotetramerization of the phthalonitrile derivative bearing 3-methoxybenzyloxy units.

Alzheimer's disease and related dementias have a multifactorial aetiology and heterogeneous biology. The current study aims to identify different biological signatures in a deeply phenotyped memory clinic patient population. In this cross-sectional study, we analysed 49 pre-specified proteins using a multiplex antibody-based suspension bead array in 278 CSF samples from the real-world research database and biobank at the Karolinska University Hospital Memory Clinic, Solna, Sweden.
