Objective: The aim was to identify whether interleukin-1 receptor antagonist (IL-1ra) inhibits interleukin-1 alpha (IL-1 alpha)-induced intercellular adhesion molecule-1 (ICAM-1) expression on cultured orbital fibroblasts, and adhesion of peripheral blood mononuclear cells (PBMC) to orbital fibroblasts, and to investigate the clinical application potential of IL-1ra in the treatment of Graves' ophthalmopathy (GO).
Methods: Cultured orbital fibroblasts from patients with GO and controls were challenged with IL-1 alpha or/and IL-1ra. Immunocytochemical staining was used to examine the changes of ICAM-1 in response to IL-1ra treatment; fluorescent photomicroscope was used to measure the adhesion between the labeled PBMC and orbital fibroblasts. Neutralizing antibody against ICAM-1 was used to demonstrate the role of ICAM-1 in the IL-1 alpha-induced adhesion.
Results: IL-1ra inhibits IL-1 alpha-induced ICAM-1 expression in cultured orbital fibroblasts both from GO patients and controls; IL-1ra inhibits IL-1 alpha-induced adhesion of PBMC to orbital fibroblasts in a concentration and time dependent manner. Moreover, a monoclonal anti-human ICAM-1 antibody produced a concentration dependent inhibition of the IL-1 alpha-induced adhesion of PBMC to the fibroblasts.
Conclusions: IL-1ra inhibits IL-1 alpha-induced ICAM-1 expression in cultured orbital fibroblasts and the adhesion of PBMC to fibroblasts. IL-1 alpha-induced ICAM-1 expression may play an important role in the adhesion process. IL-1ra may be useful in the prevention or treatment of GO.

Solitary fibrous tumor, formerly also referred to as hemangiopericytoma, is a fibroblastic mesenchymal neoplasm of the orbit that rarely metastasizes but has a propensity for local recurrence. We describe 5 cases of orbital solitary fibrous tumors that were treated with radiation. In 3 patients, radiation therapy was given in the neoadjuvant setting prior to surgery.

Radiotherapy (RT) is a mainstay of cancer treatment but is limited by tumor resistance and off-target tissue damage, often mediated by therapy-induced cellular senescence. Here, we developed a "one-two punch" nanodrug, Lipo@ABT263@Au, that integrated a senolytic agent (ABT-263) with a gold-shelled liposome for radiosensitization and sustained drug release. High-throughput screening and transcriptomic analysis identified senescence as a key RT-induced vulnerability.

Graves' orbitopathy (GO) is characterized by orbital inflammatory infiltration, expansion of orbital tissues due to de novo adipogenesis and over-production of hydrophilic glycosaminoglycans, as well as myofibroblastic differentiation resulting in tissue fibrosis. Thyrotropin receptor antibody (TSH-R-Ab) is the major stimulus, which activates Thyrotropin receptor (TSH-R) / insulin-like growth factor-1 receptor (IGF-1R) and its downstream signalling in orbital fibroblasts (OF). Clinical evaluation of TSH-R-Ab, the specific biomarker of Graves' disease (GD) and the associated orbitopathy, provides important clinical information concerning diagnosis, disease monitoring and prognosis of GO.

Purpose: Thyroid-associated ophthalmopathy (TAO) is a debilitating autoimmune disorder linked to Graves' disease (GD) that is characterized by inflammation and remodeling of orbital tissues. This study focuses on the use of poly(lactic-co-glycolic) acid (PLGA)-based microspheres (MS) coupled with CD34-specific aptamers to enhance the targeted delivery of Tocilizumab (Toc), an IL-6 receptor monoclonal antibody, to CD34+ orbital fibroblasts, which is a critical cell type implicated in TAO.
Methods: The flow cytometry and aptamer-mediated pull-down assays were applied to detect the affinity of CD34 aptamers (Apts) for CD34+ orbital fibroblasts.
