Background: Asthma remains a significant public health challenge globally, particularly among older adults who face diagnostic complexity, atypical symptom profiles, and frequent comorbidities. Despite global advances in asthma control, little attention has been paid to the evolving composition and geographic disparity of modifiable risk factors in this age group.
Methods: We utilized data from the Global Burden of Disease Study 2021 to evaluate the long-term trends (1990-2021) in asthma burden attributable to high body-mass index (BMI), smoking, and occupational asthmagens among adults aged 60 years and older.
Mechanistic modeling of ion exchange chromatography (IEC) is a promising technique to improve process development. However, when considering the pH influence, model prediction becomes challenging due to the multiple pH-dependent parameters and complex interactions. In order to more effectively predict the pH gradient elution behavior, a two-step model calibration strategy was proposed for the pH-dependent steric mass action (SMA) model with the empirical correlations of characteristic charge ν and equilibrium coefficient k.

In this study, anti-Langmuirian to Langmuirian (AL-L) elution behavior was observed in cation-exchange chromatographic separation of charge variants of industrial Fc-fusion proteins. To characterize this behavior, the multi-protein Mollerup activity model was integrated into the steric mass action (SMA) model, resulting in a new model named the generalized ion-exchange (nGIEX) isotherm for multi-protein systems.

In this paper, this concept is applied to dynamic control of the collection window in the ion exchange chromatography (IEC) toward sample variations. A possible structure of a feedforward model-based control DT system was proposed.

Continuous process is a promising alternative for tradition batch process in biomanufacturing, which has higher productivity and lower material consumption. However, despite the maturation of necessary technologies for continuous process, there are few discussion about optimization of full continuous process. One possible reason is that the continuous process is such a complex and interacted process that the traditional experiment-based optimization approach is not sufficient anymore.

However, process development is difficult due to process complexity. In this work, some general and convenient guidances for three-column periodic counter-current chromatography (3C-PCC) were developed.
