CRISPR-Cas9 and Cas12a are widely used for genome editing, but their large size limits delivery efficiency. Compact Cas12f proteins offer delivery advantages but suffer from low activity. To address this limitation, we engineered an enhanced Cas12f system (exoCasMINI) by fusing T5 exonuclease to CasMINI, achieving 1.

However, the role of tetrahedrally occupied Co (Co) and Co in an octahedral site (Co) in controlling the catalytic activity remains controversial. Herein, we investigated a geometrical-site-dependent catalytic activation of ozone respectively on the Co and Co sites.

In this study, we demonstrate that homoharringtonine (HHT), an FDA-approved therapeutic agent for chronic myeloid leukaemia (CML), inhibits this immune evasion by targeting the FTO/m6A/LILRB4 signalling pathway in monocytic AML. Utilising RNA sequencing (RNA-seq) and various functional assays, we reveal that HHT treatment significantly reduces LILRB4 expression at both the RNA and protein levels, suggesting that the effects of HHT on LILRB4 are distinct from its well-established role as a protein synthesis inhibitor.

However, their applications are currently limited due to modest gene-editing activity. Here we introduce an engineered compact CRISPR-Cas12f (hpCasMINI, 554 aa), with hyper editing efficiency in mammalian cells via adding an α-helix structure to the N-terminus of an Un1Cas12f1 variant CasMINI (529 aa).
