Community-associated methicillin-resistant strains often demonstrate enhanced virulence, with different strains eliciting varied immune responses in humans. The factors that enable some strains to evade killing by immune cells remain unclear. Here, we describe studies designed to understand intracellular survival assays that revealed that while a well-characterized MRSA ST8 strain (USA300) was susceptible to phagocytic clearance, a genotypically similar MRSA ST88 strain (LVP-7) effectively evaded neutrophil killing.

Paralogs of the bifunctional nuclease, Ribonuclease J (RNase J), demonstrate varied catalytic efficiencies despite extensive sequence and structural similarity. Of the two Staphylococcus aureus RNase J paralogues, RNase J1 is substantially more active than RNase J2. Mutational analysis of active site residues revealed that only H80 and E166 were critical for nuclease activity.

Unlabelled: Extracytoplasmic function (ECF) σ factors selectively upregulate expression of specific genes in bacteria. These σ factors, belonging to the σ family, are much smaller than the primary, housekeeping σ factor with two helical domains that interact with the Pribnow box and the -35 element of the promoter DNA. Structural studies reveal that promoter specificity in a σ factor is determined by the interactions between a loop (L3) and the Pribnow box element.

Mycobacterium tuberculosis uracil-DNA glycosylase (MtUng), a key DNA repair enzyme, represents an attractive target for the design of new antimycobacterial agents. However, only a limited number of weak MtUng inhibitors are reported, primarily based on the uracil ring, and hence, lack diversity.
