The survival rates for children with cancer have increased appreciably over the last few decades; however, childhood cancer survivors continue to suffer from long-lasting sequelae. Studies have demonstrated that the presence of malnutrition, over- and under-nutrition, at diagnosis or the duration of malnutrition during treatment is associated with increased toxicity, infection, and inferior survival. Dietary habits, along with behavioral and socioeconomic status, are known factors that lead to obesity or undernutrition and can affect the prognosis and quality of life of children with cancer.

Chronic kidney disease (CKD) affects almost 10% of the global population and is a significant health issue. The presence of CKD increases the risk of fatal and non-fatal cardiovascular events, overall mortality, and progression of renal damage leading to kidney failure. To enhance the quality of real-world external comparative studies, it is essential to systematically identify, prespecify, and account for prognostic variables and effect measure modifiers (EMMs), especially between emulated target trial and real-world control arms. These factors can then be utilized to evaluate cohort comparability and perform covariate adjustments, such as in propensity score models. A systematic literature review (SLR)-based identification of prognostic factors, coupled with expert clinical review, offers a comprehensive approach to evaluating and ranking the level of evidence, while also assisting in selection of prognostic factors to assess imbalances between cohorts in single-arm trials and real-world data studies.

While CD19-directed CAR T-cell therapy represents a transformative immunotherapy for relapsed/refractory large B-cell lymphoma (r/r LBCL), more than 50% of patients ultimately progress or relapse. In the randomized cohort of the international phase-III FORUM trial, which showed the superiority of total-body irradiation (TBI) over chemotherapy-based conditioning prior to hematopoietic stem cell transplantation (HSCT) for pediatric acute lymphoblastic leukemia (ALL), type of conditioning and remission phase, but not pre-HSCT minimal residual disease (MRD), were associated with outcome. The cumulative impact of baseline comorbidities on outcomes of chimeric antigen receptor T cell (CAR-T) therapy is not well-established.
