Article Abstract

Bicyclo[2.2.1]heptanes (BCHeps) are essential polycyclic compounds in many natural products and pharmaceutical molecules. Efficient synthetic strategies for BCHeps and their derivatives are desirable and challenging. In this work, bicyclo[2.1.0]pentanes (BCPs) have been strategically employed as building blocks for the development of a [2π + 2σ] cycloaddition reaction with alkenes. The key to success is using a pyridine-assisted boronyl radical catalysis approach. This provides a modular and atom-economical method, using simple and inexpensive catalysts, for efficient synthesis of BCHeps. The substrate scope has been explored. A possible reaction mechanism has been proposed, and the synthetic utility has been demonstrated by downstream transformations.

2-Aminoacrylate (2AA) is a short-lived enamine generated as a catalytic intermediate in the dehydration of serine by serine/threonine dehydratase enzymes. 2AA is a metabolic stressor capable of inactivating important pyridoxal phosphate dependent enzymes in a cell. The aim of this study is to investigate how Adaptive Torque Control motion influences the shaping efficiency of Neolix EDMax (Neolix SAS, Évron, France) and its ability to reach working length with or without a pre-existing glide path. A total of 90 endo training blocks with an S-shape curvature were divided into three groups based on the kinematics and preparation phase: the control group, where the Neolix EDMax (Neolix SAS, Évron, France) was used for shaping after a glide path was established; the no glide path group, where the Neolix EDMax (Neolix SAS, Évron, France) was used for shaping without a glide path; and the Adaptive Torque Control group, where the Neolix EDMax (Neolix SAS, Évron, France) was used for shaping without a glide path but in an Adaptive Torque Control motion. The time for shaping, the instrument passes, and the ability to reach working length were recorded and analyzed using a one-way Anova and Tukey's HSD post hoc test.

In the absence of the broadly conserved deaminase RidA (Reactive intermediate deaminase A), and other organisms accumulate the reactive enamine species 2-aminoacrylate (2AA). Free 2AA, generated from serine by the serine/threonine dehydratase IlvA, reacts with and covalently inactivates a subset of pyridoxal 5'-phosphate-dependent enzymes. The metabolic stress caused by 2AA generates growth defects in , including (i) when l-alanine is used as a nitrogen source, (ii) when pyruvate is used as a carbon source or (iii) in the presence of exogenous serine.
