In recent years, DNA engineering technology has undergone significant advancements, with clustered regularly interspaced short palindromic repeats (CRISPR)-based target-specific DNA insertion emerging as one of the most rapidly expanding and widely studied approaches. Traditional DNA insertion technologies employing recombinases typically involve introducing foreign DNA into genes in vivo by either pre-engineering recognition sequences specific to the recombinase or through genetic crossing to incorporate the requisite recognition sequence into the target gene. However, CRISPR-based gene insertion technologies have advanced to streamline this engineering process by combining the CRISPR-Cas module with recombinase enzymes. This process enables accurate and efficient one-step insertion of foreign DNA into the target gene in vivo. Here we provide an overview of the latest developments in CRISPR-based gene insertion technologies and discusses their potential future applications.

The donor/acceptor (D/A) interfaces in bulk heterojunction (BHJ) organic solar cells (OSCs) critically govern exciton dissociation and molecular diffusion, determining both efficiency and stability. Herein, we design a double-cable conjugated polymer, SC-1F, to insert into a physically-blended D/A system to optimize the interface. We have found that SC-1F spontaneously segregates to the interface through favorable miscibility and heterogeneous nucleation with the acceptor.

Organ-on-chip (OOC) technologies, also called microphysiological systems (MPS), offer dynamic microenvironments that improve upon static culture systems, yet widespread adoption has been hindered by fabrication complexity, reliance on polydimethylsiloxane (PDMS), and limited modularity. Here, a modular MPS platform is presented, designed for ease of use, reproducibility, and broad applicability. The system comprises layered elastomeric inserts for dual monolayer cell culture, which is clamped within a reusable acrylic cassette for perfusion studies.

Introduction: The modification of an inhaler's air flow resistance influences a patient's inhalation flow profile, thereby affecting the exit velocity of an aerosol leaving the Respimat® mouthpiece. A slower inhalation maneuver results in reduced plume velocity and thus a decreased oropharyngeal deposition due to reduced impaction. This could not only lead to fewer unwanted side effects associated with inhaled therapies, but also enhance lung deposition.

Purpose: Tumor comprehensive genomic profiling (CGP) may detect potential germline pathogenic/likely pathogenic (P/LP) alterations as secondary findings. We analyzed the frequency of potentially germline variants and large rearrangements (LRs) in the RATIONAL study, an Italian multicenter, observational clinical trial that collects next-generation sequencing-based tumor profiling data, and evaluated how these findings were managed by the enrolling centers.
Patients And Methods: Patients prospectively enrolled in the pathway-B of the RATIONAL study and undergoing CGP with the FoundationOne CDx assays were included in the analysis.

The scope of this study was to analyze the racial inequalities present in the narratives of people whose family members died from COVID-19 in Brazil. A qualitative approach was adopted, which is inserted in the social constructionist perspective. Narratives about illness and death were produced through in-depth interviews with 35 subjects.
