Article Abstract

Background/aim: Cancer development involves complex interactions between immune mechanisms and the tumor microenvironment, with regulatory T cells (Tregs) being implicated in suppressing anti-tumor immunity. The X-linked gene Forkhead Box P3 (), which regulates Tregs' function, and its promoter variant rs3761548 C>A have been widely studied for their role in tumorigenesis. This meta-analysis aims to re-evaluate the association between rs3761548 and cancer risk using two statistical approaches to account for sex-based genotypic differences and X-chromosome statistical challenges.

Materials And Methods: A systematic search of PubMed, Google Scholar, and Scopus identified seventeen eligible case-control studies, including 6719 cancer patients and 6879 healthy controls. The statistical analyses employed an X-linked Generalized Linear Regression Model (GLRM) and a sex-stratified meta-analysis calculating odds ratios with 95% confidence intervals.

Results: The results indicated that the presence of the A allele increases cancer risk in White and Asian populations. Furthermore, the A allele and the AA genotype were associated with breast cancer in Asians, while the AC genotype appeared protective against acute lymphoblastic leukemia in Whites possibly through non-random X-chromosome inactivation. The GLRM proved more robust, identifying additional associations and accounting for risk factors.

Conclusion: The present study suggests a new statistical approach for analyzing X-linked genotypic data. Additionally, it underscores that the gene could be either oncogenic or tumor suppressor depending on ethnicity, cancer type and genotype distribution among the studied groups since its expression is subject to epigenetic changes related to the rs3761548 C allele.

Allogeneic hematopoietic stem cell transplantation (allo-HSCT) offers a potential cure for hematologic malignancies, but post-transplant complications, particularly graft--host disease (GVHD), remain major challenges. Regulatory T cells and interleukin-10 () play key roles in immune regulation. This retrospective study investigated the impact of donor -3279 (rs3761548) and -819 (rs1800871) single-nucleotide polymorphisms (SNPs) on clinical outcomes in 92 allo-HSCT recipients, with emphasis on umbilical cord blood transplantation (UCBT).

PD-L1, PD-1, FOXP3, and miR-155 are emerging as key modulators of immune evasion and progression of oral squamous cell carcinoma (OSCC). This study investigated the clinical relevance of their gene expression and variants in HPV-negative OSCC. Bulk-tissue mRNA expression was evaluated in 70 patients, while variants in (rs36084323), (rs822336, rs4143815, copy number variation), (rs3761548, rs2232365), and (rs767649) were assessed in 134 patients.

Gene polymorphisms associated with immunosuppression and their implications in the onset of cervical cancer.

Women Health

July 2025

Laboratory of Molecular Genetics and Immunology, Department of Immunology, Parasitology and General Pathology, Center of Biological Sciences, State University of Londrina, Londrina, Paraná, Brazil.

Unlabelled: The immune system plays a crucial role in determining the resolution or development of cervical pre-malignant lesions that culminate in cervical cancer (CC). A microenvironment with low inflammatory patterns is often associated with a worse prognosis.

Aim: To evaluate polymorphisms rs3087465 in the and rs3761548 in the genes, as well as sociodemographic, reproductive, and sexual behavior data which may be associated with CC development.

FOXP3 gene polymorphisms are associated with indeterminate clinical form of Chagas disease.

The forkhead box protein 3 (FOXP3) transcription factor is the main marker of regulatory T-cell (Treg) development and activation, a subpopulation involved in immune system regulation, self-tolerance, and protection against infections. We previously showed that Treg cells control the exacerbated immune response and morbidity in chronic Chagas disease, by modulating the cytokine environment and killing effector cells. Although FOXP3 gene polymorphisms have already been studied in several diseases, their role in Chagas disease is underreported.
