Methylglyoxal (MGO), a reactive glycolytic byproduct, contributes to skeletal muscle atrophy by promoting oxidative stress, inflammation, and protein degradation. This study investigated the therapeutic potential of -resveratrol and hesperidin (tRES-HESP) supplementation, alone or in combination with aerobic exercise, in ameliorating MGO-induced muscle dysfunction in mice. ICR mice were divided into five groups and treated with MGO, tRES-HESP, treadmill exercise, or both interventions over eight weeks. Muscle mass, grip strength, endurance performance, histopathology, and molecular biomarkers were assessed. Combined treatment with tRES-HESP and exercise significantly improved muscle function and morphology, restored muscle mass, and suppressed fibrosis. In terms of signaling pathways, the protective effects were associated with activation of the AMPK/SIRT1/PGC-1α pathway for mitochondrial biogenesis, stimulation of the AKT/mTOR pathway for muscle protein synthesis, suppression of MuRF1-mediated protein degradation, and upregulation of MyoD and MyHC, key markers of muscle regeneration and structural integrity. Additionally, inflammatory balance was restored via suppression of pro-inflammatory IL-6 and elevation of anti-inflammatory IL-10 levels. These findings highlight the synergistic benefits of combining polyphenol supplementation with aerobic exercise as a promising strategy for mitigating muscle atrophy under metabolic stress conditions.

Maintenance of organismal function requires tightly regulated biomolecular communication. However, with aging, communication deteriorates, thereby disrupting effective information flow. Using information theory applied to skeletal muscle single cell RNA-seq data from young, middle-aged, and aged animals, we quantified the loss of communication efficiency over time.

Objectives: To explore the relationships between cardiac parameters and body composition indices, identifying predictors of subclinical cardiac systolic dysfunction.
Methods: Using anthropometric and serological parameters, echocardiography, and body composition analysis, this study evaluated metabolic profiles, cardiac remodeling patterns, and body composition characteristics in young adult obese patients, while quantifying the correlations between cardiac parameters and body composition indices. Subclinical left ventricular systolic dysfunction was defined as global longitudinal strain (GLS) < 18%.

Introduction: No head-to-head studies comparing the efficacy of avalglucosidase alfa (AVA) with cipaglucosidase alfa + miglustat (Cipa+mig) have been conducted in patients with late-onset Pompe disease (LOPD). Two indirect treatment comparisons (ITCs) were conducted to estimate the effects of AVA versus Cipa+mig.
Methods: ITCs were conducted using simulated treatment comparisons (STCs), adjusting for differences in prognostic factors and treatment effect modifiers.

The explanation for how acutely stressful experiences could result in proximal health outcomes has been lacking in occupational health research. Although scholars have argued that individual personality and affect could worsen health behaviors, we believe that these qualities also could intensify the experience of acute stressors, potentially explaining why acutely stress encounters result in poor health outcomes for some people, but not others. Our study examines three individual differences - worry, negative affect, and positive affect - that are relevant to differential stress anticipation, reactivity, and recovery.

Background: Electrical impedance myography (EIM) has been proposed as an efficient, non-invasive biomarker of muscle composition in facioscapulohumeral muscular dystrophy (FSHD).
Objective: We investigate whether EIM parameters are associated with muscle structure measured by magnetic resonance imaging (MRI), muscle histology, and transcriptomic analysis as well as strength at the individual leg muscle level.
Methods: We performed a multi-center cross-sectional study enrolling 33 patients with FSHD.
