Article Abstract

Gray mold disease, caused by , severely impacts grape production worldwide. Although proanthocyanidins (PAs) contribute to fungal pathogen resistance, their role in grape defense against remains unclear. Here, we demonstrate that VvMYBPA1, a key transcriptional regulator of PA biosynthesis, negatively modulates resistance in grape berries. While infection suppressed endogenous VvMYBPA1, its agroinfiltration-mediated transient overexpression in berries elevated susceptibility, paralleling reduced β-1,3-glucanase (BGL) and polyphenol oxidase (PPO) activities. Additionally, VvMYBPA1 overexpression elevated expression and reduced peroxidase (POD) activity, resulting in excessive hydrogen peroxide (HO) accumulation and more cell death. Our results reveal that VvMYBPA1 negatively regulates resistance by disrupting antioxidant enzyme activity and ROS homeostasis, providing new insights into the interplay between PA biosynthesis and fungal defense mechanisms.

Background And Hypothesis: Schizophrenia is linked to hippocampal dysfunction and microglial inflammatory activation. Our prior clinical findings revealed significantly reduced transient receptor potential vanilloid 1 (TRPV1) expression in both first-episode and recurrent schizophrenia patients, with levels inversely correlating with symptom severity, implicating TRPV1 dysfunction in disease progression. Preclinical maternal separation (MS) models recapitulate schizophrenia-like behavioral and synaptic deficits, paralleled by hippocampal microglial TRPV1 downregulation.

Unlabelled: Cholesterol 25-hydroxylase (CH25H), an interferon-stimulated gene (ISG), has been implicated in broad-spectrum antiviral immunity. Here, we identify CH25H as a potent suppressor of hepatitis B virus (HBV) replication that significantly outperforms IFN-α in reducing HBV DNA, pregenomic RNA (pgRNA), HBsAg, and HBeAg, without inducing cytotoxicity. However, CH25H is weakly expressed in hepatocytes and only modestly induced by type I interferon.

This study investigated the effects of a low-frequency polarized electric field (LFPEF) on postharvest disease resistance and storage quality of grapes. LFPEF treatment (3 h/d) significantly reduced weight loss, suppressed lesion expansion, and maintained fruit firmness by reinforcing cell wall integrity and enhancing defense-related enzyme activities. Mechanistic analyses indicated that LFPEF activated Ca signaling, promoted calcium accumulation, and upregulated calcium sensor genes, thereby contributing to membrane stabilization.

Membrane-protein quality control in Escherichia coli involves coordinated actions of the AAA+ protease FtsH, the insertase YidC and the regulatory complex HflKC. These systems maintain proteostasis by facilitating membrane-protein insertion, folding and degradation. To gain structural insights into a putative complex formed by FtsH and YidC, we performed single-particle cryogenic electron microscopy on detergent-solubilized membrane samples, from which FtsH and YidC were purified using Ni-NTA affinity and size-exclusion chromatography.
