A hallmark of Alzheimer's disease (AD) is the accumulation of extracellular amyloid-β plaques in the brain. Amyloid-β is a 40-42 amino acid peptide generated by proteolytic processing of amyloid precursor protein (APP) via membrane-bound proteases. APP is a transmembrane protein, and its trafficking to sites of proteolysis represents a rate-limiting step in AD progression. Although APP processing has been well-studied, its trafficking itinerary and machinery remain incompletely understood. To address this, we performed an unbiased interaction screen for interactors of the APP cytosolic tail. We identified previously characterised APP binders as well as novel interactors, including RABGAP1. We demonstrated that RABGAP1 partially co-localises with APP and directly interacts with a YENPTY motif in the APP cytosolic tail. Depletion or overexpression of RABGAP1 caused mistrafficking and misprocessing of endogenous APP in human and rodent neurons. This effect is dependent on the GAP activity of RABGAP1, demonstrating that RABGAP1 affects the trafficking of APP by modulating RAB activity on endosomal subdomains. This novel trafficking mechanism has implications for other NPXY cargoes and presents a possible therapeutic avenue to explore.

Academic-Practice Partnerships (APPs) are formal relationships between academic institutions and healthcare organizations that aim to improve healthcare delivery by preparing practice-ready nurses who readily transition into the workforce. The purpose of this article is to describe the intentional process used to create a new APP between a healthcare organization in a rural setting with a medium sized Midwestern university. The focus of the APP is to enhance leadership experiences for prelicensure students from the academic setting and make the meaningful work and impact of the practice partner nurse leaders visible to students.

Under physiological conditions, amyloid precursor protein (APP) is critically important for normal brain development, neurogenesis, neuronal survival, and synaptic signaling. Dyshomeostasis of APP increases deposition and accumulation of amyloid β (Aβ) in the brain parenchyma and cerebral blood vessels thereby leading to development of Alzheimer's disease and cerebral amyloid angiopathy. In this review, we critically examine existing literature supporting the concept that endothelial APP performs important vascular protective functions in the brain.

The flammability and poor ultraviolet (UV) aging resistance of polylactic acid (PLA) limit its applications outdoors and in fields requiring flame retardancy. To address these limitations, this study designed ammonium polyphosphate (APP) as the core, the biopolymer chitosan (CS) as the inner shell, and lignin (LK) as the outer shell. CS and LK are deposited on the surface of APP via electrostatic interaction in the aqueous phase to prepare a core-shell structure flame retardant APP@CS@LK with anti-UV aging properties.
