has emerged as a fungal pathogen of particular concern owing in part to its propensity to exhibit antifungal resistance, especially to the commonly prescribed antifungal fluconazole. A mutation in , which encodes a zinc cluster transcription factor, has been shown to confer increased resistance to fluconazole. In this work, we aimed to determine how mutations in exert this effect. mutations leading to A640V, A657V, and F862_N866del, found in fluconazole-resistant clinical isolates, were introduced into two susceptible Clade I backgrounds using CRISPR-Cas9 gene editing. These mutations conferred increased fluconazole resistance, as well as increased resistance to other triazoles as measured by broth microdilution. RNA-seq revealed that the ATP-binding cassette (ABC) transporter gene as well as the major facilitator superfamily (MFS) transporter gene were both upregulated in the presence of these mutations. Disruption of increased susceptibility in strains with mutations, whereas disruption of had little to no effect. However, disruption of both and resulted in an additional increase in susceptibility as compared with disruption of alone. mutations, leading to A640V, A657V, and F862_N866del all result in increased resistance to fluconazole and other triazole antifungals and increased expression of both and in . Together, these data suggest is the primary driver of resistance conferred by these mutations.

In this study, employing a 2D electrodeposition in situ assembly method, a high-performance HS sensor based on a p-n type CuO-CuFeO heterostructure ordered nanowire arrays was successfully fabricated on silicon substrates. Compared to CuO, CuO-CuFeO nanowire arrays exhibits an ideal interfacial barrier structure and higher initial resistance, with a response to 10 ppm of HS at room temperature (20 ± 3 °C) increased by 225 times and a response time reduced by over 2400 s. Study Question: Does weight loss from a hypocaloric dietary intervention improve antral follicle dynamics in women with PCOS?
Summary Answer: During a 3-month hypocaloric dietary intervention, women with PCOS who experienced clinically meaningful weight loss showed more organized antral follicle development including fewer recruitment events, but no change in the overall frequency of selection, dominance, or ovulation.
What Is Known Already: There is a spectrum of disordered antral follicle development in women with PCOS including excessive follicle recruitment and turnover, decreased frequency of selection and dominance, and failure of ovulation. Lifestyle intervention aimed at weight loss is recommended to improve metabolic health in women with PCOS yet benefits on ovarian follicle development and ovulation are unclear.

Protein translation regulation is critical for cellular responses and development, yet how elongation stage disruptions shape these processes remains incompletely understood. Here, we identify a single amino acid substitution (P55Q) in the ribosomal protein RPL-36A of Caenorhabditis elegans that confers complete resistance to the elongation inhibitor cycloheximide (CHX). Heterozygous animals carrying both wild-type RPL-36A and RPL-36A(P55Q) develop normally but show intermediate CHX resistance, indicating a partial dominant effect.

We show that the ground state of a weakly charged two-dimensional electron-hole fluid in a strong magnetic field is a broken translation symmetry state with interpenetrating lattices of localized vortices and antivortices in the electron-hole-pair field. The vortices and antivortices carry fractional charges of equal sign but unequal magnitude and have a honeycomb-lattice structure that contrasts with the triangular lattices of superconducting electron-electron-pair vortex lattices. We predict that increasing charge density or a weakening magnetic field drives a vortex delocalization transition that would be signaled experimentally by an abrupt increase in counterflow transport resistance.
