Article Abstract

In brains of individuals who had sustained repetitive head trauma, advanced pathologic tau protein in neurons and axons within temporal cortices followed patterns seen in homologous cortico-cortical connections in nonhuman primates. The relational Structural Model, which is based on the universal principle of the systematic variation of cortical laminar structure, has successfully predicted the relative laminar distribution of cortico-cortical connections based on the relative similarity/difference in laminar structure in pairs of linked areas. Here, the Structural Model successfully predicted the graded laminar distribution and density of pathologic tau in chronic traumatic encephalopathy and was validated by a computational progression model. By contrast, early and sporadic tau pathology in the depths of sulci in chronic traumatic encephalopathy followed local columnar connectivity rules. These findings support applicability of a theoretical model to unravel the direction and progression of tau pathology in neurodegeneration via cortical connection mechanisms. Cortical pathways converging on temporal cortices may help explain the inexorable spread of pathologic tau to widespread cortical areas accompanied by decline in emotional and cognitive processes in humans with repetitive head trauma.

Traumatic brain injuries (TBIs) are a risk factor for Alzheimer's disease (AD), and share several important pathological features including the development of neurofibrillary tangles (NFT) of tau protein. While this association is well established, the underlying pathogenesis is poorly defined and current treatment options remain limited, necessitating novel methods and approaches. In response we developed "TBI-on-a-chip", an trauma model utilizing murine cortical networks on microelectrode arrays (MEAs), capable of reproducing clinically relevant impact injuries while providing simultaneous morphological and electrophysiological readout.

Mitochondrial dysfunction is a shared hallmark of neurodegenerative disorders, including Alzheimer's disease (AD) and tauopathies among others. Pathological alterations of the microtubule-associated protein Tau can disrupt mitochondrial dynamics, transport, and function, ultimately leading to neuronal toxicity and synaptic deficits. Understanding these processes is crucial for developing therapeutic interventions.

Anti-IgLON5 disease is an autoimmune encephalitis with more chronic presentation including memory decline, sleep disorder, bulbar symptoms and movement disorder. Post-mortem brains of patients with anti-IgLON5 disease show neurodegeneration with tau deposition sparking interest in this 'acquired tauopathy' as a disease model for neurodegeneration, yet mechanisms of neurodegeneration remain unknown. Using a reductionist human iPSC-derived neuron-antibody model, we applied proteomics approach, electrophysiology and live cell imaging.

BackgroundAlzheimer's disease (AD) is the most common neurodegenerative disorder. While AD diagnosis traditionally relies on clinical criteria, recent trends favor a precise biological definition. Existing biomarkers efficiently detect AD pathology but inadequately reflect the extent of cognitive impairment or disease heterogeneity.

Background: As a common postoperative neurological complication, postoperative delirium (POD) can lead to poor postoperative recovery in patients, prolonged hospitalization, and even increased mortality. However, POD's mechanism remains undefined and there are no reliable molecular markers of POD to date. The present work examined the associations of cerebrospinal fluid (CSF) sTREM2 with CSF POD biomarkers, and investigated whether the effects of CSF sTREM2 on POD were modulated by the core pathological indexes of POD (Aβ42, tau, and ptau).
