Inbred lines of , a wild relative of cultivated watermelon, are widely used as rootstocks to control soil-borne diseases for watermelon ( ) production. The most commonly used rootstock, 'Carolina strongback' (Syngenta, Basel, Switzerland) flowers weeks later than commercial watermelon cultivars, which delays the onset of female flowering (DFF) of the scion, leading to an undesirable delay in fruit maturity and harvesting. Understanding the genetics of DFF in a population will facilitate the development of rootstocks with the early flowering habits preferred for commercial production.

Adeno-associated virus (AAV) gene therapy is often limited by pre-existing neutralizing antibodies (NAbs), yet current assays for NAb detection lack standardization and rarely quantify uncertainty, complicating cross-study comparisons. We present coreTIA (core Transduction Inhibition Assay), a comprehensive framework providing a modular experimental protocol and a statistically robust analysis pipeline. This integrated method delivers precise, reproducible NAb titers with quantified uncertainty for every result.

Background: Subcutaneous immunoglobulin (SCIg) is a promising alternative to intravenous Ig (IVIg) for the treatment of idiopathic inflammatory myositis (IIM), thanks to its more favorable safety profile, reduced costs, and lower impact on patients' quality of life. We assessed the short- and long-term effectiveness and safety of recombinant human hyaluronidase-facilitated SCIg (hf-SCIg) in patients with IIM treated at different referral centers in Italy.
Methods: A multicenter, retrospective, real-life cohort study was conducted on consecutive adult patients diagnosed with IIM according to the EULAR/ACR criteria, treated with hf-SCIg for remission induction or maintenance.
Gaucher disease type 1 is a lysosomal storage disorder caused by mutations that reduce glucocerebrosidase activity, leading to glycolipid buildup, particularly in macrophages. To develop a curative approach, we established a high-efficiency genome editing platform for human and murine hematopoietic stem-progenitor cells using CRISPR/Cas9, recombinant adeno-associated virus serotype 6. To enhance homology-directed DNA repair while minimizing genotoxicity, we incorporated a new 53BP1 inhibitor, a ubiquitin variant that promotes DNA end resection and significantly increases editing efficiency.

Juvenile nephronophthisis (NPHP) is the most common genetic cause of pediatric chronic kidney disease (CKD). Its nonspecific findings such as intermittent fatigue, nausea, or vomiting, often delay diagnosis, especially without extra-renal manifestations. This case study reports a 9-year-old boy with a week of acute-on-chronic vomiting, a year of nausea and fatigue, and new onset polydipsia and nocturnal enuresis.
