Article Abstract

Insects have a powerful olfactory system that is far more selective and sensitive than artificial detectors. Insect odorant receptors (ORs) are key components of the system, which are ligand-gated ion channels comprising a specific odorant-sensing OR and a highly conserved odorant receptor co-receptor (Orco). However, the stoichiometric ratios of the heterotetramers remain inconclusive, and the molecular mechanism by which the ligand initiates channel opening is still not fully understood. The present study is based on the technical approach of molecular dynamics (MD) simulation. We predict the spatial structures of locust LmOR35-Orco heterotetramer under various stoichiometric ratios, construct it within a membrane environment, and compare the structural changes of LmOR35-Orco before and after ligand binding. Furthermore, we analyze the molecular mechanism of LmOR35-Orco across different architectures. Our findings propose an optimal architecture (1OR:3Orco) for insect heteromeric odorant receptors, elucidate the molecular mechanism underlying receptor activation due to ligand-induced ion channel opening, and identify critical residues involved in ligand recognition and ion channel gating. This study provides valuable insights into the regulatory mechanism of insect olfaction and has significant implications for function modification and the development of bionic electronic noses.

Embryo transfer (ET) is a valuable reproductive technology in pigs, albeit its efficiency remains significantly lower than that of natural mating or artificial insemination (AI), owing to high embryonic death rates. Pomegranate (Punica granatum L) is a rich source of bioactive compounds, including punicalagin, ellagic acid, anthocyanins, and urolithins, which contribute to its broad pharmacological potential. This review summarizes evidence from in vitro and in vivo experiments, as well as clinical studies, highlighting pomegranate's therapeutic effects in inflammation, metabolic disorders, cancer, cardiovascular disease, neurodegeneration, microbial infections, and skin conditions. Mechanistic insights show modulation of pathways such as nuclear factor-kappa B (NF-κB), mitogen-activated protein kinase (MAPK), alpha serine/threonine-protein kinase (AKT1), and nuclear factor erythroid 2-related factor 2 (Nrf2).

Parasitoid wasps are a diverse group of insects with a unique parasitic lifestyle that allows them to spend their lives closely interacting with their insect hosts, facilitated by parasitic effectors, including venom, polydnaviruses, and teratocytes. These effectors manipulate various aspects of insect host biology to increase the survival of the parasitoids' offspring. During the last two decades, omics and functional studies have significantly advanced our understanding of how parasitoids manipulate their hosts at the molecular level.
