Article Abstract

Chemical biopsy by solid-phase microextraction (SPME) with sorbent-coated fibers offers monitoring of biological processes in a significantly less invasive manner compared to conventional tissue biopsy. The developed device features a self-protective design by using an acupuncture needle coated with biocompatible material only in a recessed section. The coating comprises micron-sized naturally wettable sorbent particles embedded in a durable, biocompatible binder, ensuring broad analyte extraction coverage without the need for solvent activation. This design allows professionals in biochemical research and medical staff to use the device for in vivo monitoring of tissue concentrations of endogenous and/or exogenous substances without introducing activation solvent to the investigated system. The device is successfully used to sample anti-cancer drugs in both animal models and human patients undergoing in vivo lung perfusion (IVLP) surgery, and then determine the drug concentration level by LC/MS. Finally, a proof-of-concept experiment using a microfluidic open interface (MOI) to directly desorb and introduce the extract analytes from the coating to MS detection is proposed for potential on-site real-time drug monitoring during the surgery.

This review analyzes Russian and international literature on the treatment of bilateral limbal stem cell deficiency (LSCD), focusing on the use of Simple Oral Mucosal Epithelial Transplantation (SOMET) as a surgical method for restoring the ocular surface. Contemporary sources report 64 cases of SOMET used in the treatment of bilateral LSCD: 35 cases of chemical burns, 16 of thermal burns, 7 cases of Stevens-Johnson syndrome, 1 keratitis, 1 cicatricial pemphigoid, 1 dermoid, 1 case of drug-induced LSCD (mitomycin C), etc. Notably, all transplantations resulted in complete epithelialization, and in 3 cases, penetrating keratoplasty was subsequently performed with favorable functional and anatomical outcomes.

Drug-induced hepatotoxicity (DIH), characterized by diverse phenotypes and complex mechanisms, remains a critical challenge in drug discovery. To systematically decode this diversity and complexity, we propose a multi-dimensional computational framework integrating molecular structure analysis with disease pathogenesis exploration, focusing on drug-induced intrahepatic cholestasis (DIIC) as a representative DIH subtype. The epidermal growth factor receptor (EGFR) is a common diver gene for lung cancer (NSCLC), which leads to an increasing death rate worldwide. This study reports the design, synthesis, and biological evaluation of triazole-clubbed pyrimidine derivatives (RDa-RDm) as potential anticancer agents.
