Neurons and their subcellular compartments exhibit distinct forms of excitability. In 1948, Alan Hodgkin described different classes of neuronal excitability, each characterized by unique spiking responses to a constant stimulus. Despite these early insights, the mechanisms by which membrane properties influence spike initiation and excitability remain poorly understood. This review explores the nonlinear dynamics underlying spike initiation across excitability classes, emphasizing how these differences contribute to the neural encoding and processing of diverse information. Within a single neuron, compartments such as the soma, axon initial segment (AIS), and axon can exhibit functionally distinct excitability profiles due to differences in ion channel expression and membrane properties. For instance, the biophysical properties of myelinated axons, particularly the expression and distribution of voltage-gated potassium (Kv1) channels, play a key role in maintaining the directional fidelity of action potential propagation by facilitating orthodromic transmission and suppressing antidromic activity. These compartment-specific dynamics underscore the intricate design of neural systems to maintain the precision and efficiency of neural signalling. Moreover, perturbations in excitability are implicated in various neurological disorders, including epilepsy and chronic pain, highlighting the importance of maintaining physiological excitability profiles. By exploring these mechanisms, this review aims to provide insight into how alterations in membrane biophysics may inform future therapeutic strategies targeting excitability-related pathologies.

Introduction: Immune-deficient/disordered people (IDP) elicit a less robust immune response to COVID-19 vaccination than the general US population. Despite millions of IDP at presumed elevated risk, few population-level studies of IDP have been conducted in the Omicron era to evaluate breakthrough infection-related outcomes.
Methods: We followed a prospective cohort of 219 IDP and 63 healthy volunteers (HV) in the USA from April 2021 (Alpha variant peak) to July 2023 (Omicron XBB variant peak).

Introduction: Landau-Kleffner Syndrome (LKS) is a subtype of epileptic encephalopathy with spike-wave activation during sleep (EESWAS), characterized by acquired aphasia associated with the emergence of epileptiform abnormalities.
Patients And Methods: A cross-sectional descriptive study was conducted in a tertiary hospital, involving one group of children with LKS and another with EESWAS. The aim was to compare the clinical, neurophysiological, and neuropsychological aspects of both groups.

MicroRNAs (miRNAs) are considered more stable than mRNA, but the impact of progressive thawing of biological samples after freezing as may happen during shipping delays has not been quantified. To address this, we utilized digital PCR to estimate the absolute concentrations of select miRNAs following progressive thawing of human plasma and maintenance at ambient temperature. Specifically, we quantified let-7b-3p, miR-144-5p, miR-150-5p, miR-517a-3p, miR-524-5p, and miR-1283, which have varying abundance in plasma.

Objective: To describe observed intraocular pressure (IOP) changes following vitrectomy (PPV) surgery and PPV combined with phacoemulsification in eyes with and without glaucoma.
Methods: A total of 20,894 patients from the American Academy of Ophthalmology IRIS® Registry (Intelligent Research in Sight) who underwent vitrectomy surgery for epiretinal membrane or vitreous opacities from January 2016 to March 2023 were included. Mean IOPs from postoperative day 1, days 2-10, days 11-30, and then monthly through 6 months were compared to baseline.
African swine fever virus (ASFV) is an important transboundary animal pathogen with significant impacts on the global swine industry. Overwhelming proinflammatory responses are a major virulence mechanism for ASFV, but the dynamics of these changes during clinical disease are not completely understood.
