Long-duration spaceflight exposes astronauts to various stressors that can alter human physiology, potentially causing immediate and long-term health effects. These stressors can damage biomolecules, cells, tissues, and organs, leading to adverse outcomes. Developing adverse outcome pathways (AOPs) relevant to radiation exposure can guide research priorities and inform risk assessments of future space exploration activities.

This study introduces a novel computational approach based on ratchet-and-pawl molecular dynamics (rMD) for accurately estimating ligand dissociation kinetics in protein-ligand complexes. By integrating Kramers's theory with Bell's equation, our method systematically investigates the relationship between the effective biasing force applied during simulations and the ligand residence times. The proposed technique is demonstrated through extensive simulations of the benzamidine-trypsin complex, employing first an implicit solvent model (multi-eGO) to set up the approach parameters and then an explicit solvent model.

Aspergillus has become the second most common causative agent of invasive fungal infections and is the leading cause of death from fungal infections. English-language publications ranging from 1975 to 2022 collected from the Web of Science Core Collection database were analyzed visually using VOSviewer, R package Bibliometrix, Scimago graphic, Gephi, Pajek, and Microsoft Excel 365. Drug delivery to the central nervous system (CNS) is primarily hindered by the blood-brain barrier (BBB). To address this, mucoadhesive formulations have been designed to prolong residence time at the application site. In this study, we comprehensively characterized the physicochemical and mucoadhesive properties of hyaluronic acid tyramine (HATA) photocrosslinked hydrogels using rheological methods, nanoindentation, contact angle goniometry, and advanced confocal microscopy.
