Article Abstract

Influenza A H1N1 and Influenza B are two of the most prevalent airborne viruses, capable of triggering seasonal epidemics that affect millions of individuals globally. Rapid on-site identification of these two viruses is crucial to accurate clinical diagnosis and prompt control of public health risks. This study presents a novel one-step reverse transcription recombinase polymerase amplification (RT-RPA) method integrated with a centrifugal microfluidic chip for rapid differential diagnosis of Influenza A H1N1 and Influenza B viruses. Addressing limitations of conventional techniques, the platform merges reverse transcription and amplification into a single step, reducing aerosol contamination risk and enabling on-site detection. Specific primers and probes targeting H1N1-HA, H1N1-NA, IVB-HA, and IVB-NA genes were designed, achieving detection limits of 10 copies/mL for H1N1-HA and 10¹ copies/mL for other targets within 25 min. The system demonstrated 100% specificity against common respiratory viruses and no cross-reactivity. Validation with 26 aerosol samples collected from public areas using bioaerosol sampler identified two H1N1-positive cases, showing 96.15%-100% consistency with off-chip qRT-RPA. The chip required only 6.25 µL of sample, exhibited high reproducibility (CV <5%). This microfluidic-RT-RPA system offers a practical solution for point-of-care influenza subtyping, enhancing outbreak control and clinical decision-making in resource-limited settings.

Patient-derived cancer organoids (PDCOs) are a valuable model to recapitulate human disease in culture with important implications for drug development. However, current methods for rapidly and reproducibly assessing PDCOs are limited. Label-free imaging methods are a promising tool to measure organoid level heterogeneity and rapidly screen drug response in PDCOs.

Hyperglycemia-induced oxidative stress and inflammation critically impair diabetic bone defect repair. Here, a radially oriented microchannel scaffold (D-GSH@QZ) was developed via a directional freezing technique integrated with photo-cross-linking strategies. The scaffold was fabricated from gelatin methacryloyl, silk fibroin methacryloyl, and nanohydroxyapatite (HAp) to mimic the natural bone matrix, while incorporating quercetin-loaded ZIF-8 nanoparticles (Qu@ZIF-8) for pathological microenvironment modulation.

Drosophila seminal fluid proteins (SFPs) are often cited as an example of interlocus sexual conflict, wherein the proteins increase male fitness while decreasing female fitness, spurring recurring female counter adaptations and rapid molecular evolution. This model predicts that male-expressed genetic variation in the accessory gland, which produces seminal fluid, should generate counter-evolving genetic pathways in females, resulting in sexual coevolution. Using a trio of D.

Structure elucidation and antioxidant activity of the radical-induced oxidation products obtained from procyanidins B1 to B4.

A- and B-type procyanidins (PCs) are widely known for their health-promoting properties, such as antioxidant activity. The limited availability of reference substances represents a major challenge, resulting in a low number of systematic studies on their health benefits. In our study, the optimised 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-induced oxidation of the B-types B1 and B2 was carried out yielding the corresponding A-types A1 (1) and A2 (2), which have an additional ether bridge between C2--C7, whereas oxidation of B3 and B4 produced a six-membered spirocyclic ring system including a spiro-carbon atom at C8t (3-5).
