In previously reported electron capture dissociation (ECD) mass spectrometry of linked glycopeptides, ECD did not provide efficient backbone fragmentation between the two glycans in a single peptide, which did not allow the assignment of a correct sugar composition in each glycan. In this study, we found hydrogen bonds and proton-bound interactions between two -glycans, so the backbone fragments between the two -glycans were not observed, although the backbone was cleaved. Such behavior is typical for precursor ions produced by conventional electrospray ionization. Supercharging reagent 3-nitrobenzyl alcohol increases the precursor charge state, which leads to the protonation of each glycan, and the resulting Coulombic repulsive force is sufficient to quench the noncovalent binding. Highly charged precursors produced using the supercharged reagents yield intense c' and z fragments between the glycans in ECD analysis. Computational molecular structure calculations and differential mobility spectrometry validated the model of glycan formations in precursors.

Alstoniaschines A‒I (1‒9), nine previously alkaloids sharing five different skeletons were obtained from the leaves of Alstonia scholaris. The structures and absolute configurations were established by their extensive spectroscopic data analyses, including NMR, HRESIMS, X-ray crystallography data, and theoretical ECD calculations. Compounds 1, 2, 3, and 9 exerted significant protective effect against oxidative stress and inflammatory damage of podocytes induced by high glucose, manifesting as the increase of superoxide dismutase, catalase, glutathione peroxidase, alongside the reductions of malondialdehyde, nitric oxide, lactate dehydrogenase.

Chemical investigations of the -butanol extract of the roots of were carried out using column chromatography, flash, semi-preparative HPLC, and chiral HPLC. Five unidentified compounds, including two prenylated coumarin glucosides, two prenylated furanocoumarin glucosides, and a benzofuran glucoside, together with twelve known compounds, were isolated from the -butanol fraction of extract. The structures of these compounds were identified by HRMS, NMR, UV, ECD in combination with quantum chemical calculations, and comparison with the literature.

Dipeptidyl-peptidase (DPP)-IV inhibition by penultimate N-terminus Pro-containing peptides is a promising strategy for Type 2 diabetes (T2D) management, as it prevents the degradation of incretin hormones (DPP-IV substrates) like glucagon-like peptide-1 (GLP-1), thereby prolonging their half-life. However, the stability and bio-accessibility of these peptides are crucial to their efficacy in orally administered therapeutics. We previously identified LPCL and TPFLPDE peptides from tilapia viscera by-products hydrolysates, which exhibited significant DPP-IV inhibition in vitro and in situ while effectively preserving active GLP-1 levels after 2 h treatment in STC-1 cells under basal glucose conditions.

Pentameric ligand-gated ion channels control synaptic neurotransmission via an allosteric mechanism, whereby agonist binding induces global protein conformational changes that open an ion-conducting pore. For the proton-activated bacterial () ligand-gated ion channel (GLIC), high-resolution structures are available in multiple conformational states. We used a library of atomistic molecular dynamics (MD) simulations to study conformational changes and to perform dynamic network analysis to elucidate the communication pathways underlying the gating process.
