Two-photon light sheet microscopy offers great potential for a range of biological applications. Still, its practical implementation is impeded by the high cost of laser sources, the complexity of construction, and the challenges associated with adapting to existing microscope setups. Here, we release an open-source design that addresses these limitations by providing detailed building instructions for transforming a brightfield microscope into a versatile one- and two-photon light sheet system. Our design incorporates a specially designed broadband hollow core fiber, enabling the simultaneous utilization of visible laser alongside an expansive pulsed laser source from another setup. This integration allows for uncompromised image resolution and speed. Furthermore, the design reduces the complexity of construction, alignment, and overall cost, thereby significantly enhancing the accessibility of this technology ( https://github.com/LJPZebra/OLU ).

Background: Tissue clearing techniques combined with light-sheet fluorescence microscopy (LSFM) enable high-resolution 3D imaging of biological structures without physical sectioning. While widely used in neuroscience to determine brain architecture and connectomics, their application for spinal cord mapping remains more limited, posing challenges for studying demyelinating diseases like multiple sclerosis. Myelin visualization in cleared tissues is particularly difficult due to the lipid-removal nature of most clearing protocols, and alternative immunolabeling approaches failed to reach satisfying results.

Assembly is a natural process where molecules spontaneously form ordered structures with specific functions. View Article and Find Full Text PDFUnderstanding musculoskeletal joints from a 3D multiscale perspective, from molecular to anatomical levels, is essential for resolving the confounding relationships between structure and pain, elucidating the intricate mechanisms regulating joint health and diseases, and developing new treatment strategies. Here, we introduce a musculoskeletal joint immunostaining and clearing (MUSIC) method specifically designed to overcome key challenges of immunostaining and optical clearing of intact joints. Coupled with large-field light sheet microscopy, our approach enables 3D high-resolution, microscale neurovascular mapping within the context of whole-joint anatomy without the need for image coregistration across various joints, including temporomandibular joints, knees, and spines, and multiple species, including mouse, rat, and pig.

Organic semiconductors are very attractive photocatalysts for the production of solar fuels. However, their development is greatly plagued by limited visible light absorption and severe restriction of photoexcited charge carrier separation and transfer caused by the exciton effect resulting from inherent dielectric constraints. Herein, a three-motif molecular junction hydrogen evolution photocatalyst is constructed by linking a donor-acceptor-donor (D-A-D) molecule integrating the photosensitizer unit and the redox unit with holey carbon nitride sheets (HCNS) as a second electron acceptor unit (A) based on the covalent strategy.
