Coproporphyrin (CP)-I has emerged as a promising endogenous biomarker for investigating organic anion transporting polypeptide (OATP) 1B1/1B3-mediated drug-drug interactions. The precursor eliminated analytical (PEA) method selectively quantifies the endogenous CP-I that should be monitored, whereas the precursor oxidized analytical method quantifies the total CP-I concentration by converting all precursors into CP-I during the sample preparation process. Plasma concentrations of CP-I exhibited a greater magnitude of change in response to rifampicin treatment compared with its precursor in a previous monkey study.

Background: The DAIR procedure (debridement, antibiotic therapy, and implant retention) represents a treatment option for acute periprosthetic joint infections (PJI). Compared to revision arthroplasty, it is technically less invasive and, under appropriate indications, can preserve a well-fixed endoprosthesis. However, treatment success depends on numerous patient-, pathogen-, and procedure-related factors.

Background: The Maltalep trial in Bangladesh assessed whether single-dose rifampicin (SDR) given 8-12 weeks after bacillus Calmette-Guérin (BCG) vaccination was able to prevent excess leprosy cases due to BCG in contacts of newly diagnosed leprosy patients. After previous publication of the two years follow-up results of the trial, we now review the results after five years. Furthermore, to better understand the long-term protective effects of BCG against leprosy, we conduct post-hoc in-depth secondary statistical analyses based on the prospective interventional (randomized) Maltalep trial and a non-interventional (non-randomized) cohort study that was conducted simultaneously in the same project area.

People who smoke are at increased risk of unfavorable tuberculosis (TB) treatment outcomes compared with those who do not, but the pathways explaining this effect are unclear. We estimated the effect of smoking on a successful end-of-treatment outcome for multidrug-resistant and rifampicin-resistant (MDR/RR) TB and examined if intervening on loss to follow-up mitigates this effect. The endTB Observational Study was a prospective cohort of people with MDR/RR-TB who were treated with longer regimens containing bedaquiline and/or delamanid.
