Bifidobacterium longum plays a critical role in the human gut and exhibits diverse probiotic functions. Achieving high-density fermentation of B. longum largely depends on the composition of the culture medium and fermentation conditions. This study aimed to optimize the medium composition and fermentation parameters for B. longum HSBL001, using viable cell counts and optical density at 600 nm (OD600) as indicators. The goal was to improve biomass yield and support the development and industrial application of highly active probiotic preparations. The optimal medium composition and culture conditions were established using a combination of single-factor experiments, the Plackett-Burman design, the steepest ascent method, and the central composite design. The optimized culture medium consisted of yeast extract (19.524 g/L), yeast peptone (25.85 g/L), arginine (0.599 g/L), glucose (27.36 g/L), MnSO (0.09 g/L), MgSO (0.8 g/L), Tween-80 (1 g/L), l-cysteine hydrochloride (0.24 g/L), and methionine (0.15 g/L). The optimal culture conditions included an initial pH of 7.0, 5% inoculum size, and incubation at 37°C, yielding a final viable cell count of 4.20 × 10 colony-forming units (CFU/mL). In a 3 L bioreactor, the viable cell count reached 1.17 × 10 CFU/mL, which was 1.786 times higher than that achieved with the modified MRS medium. These findings demonstrate that the optimized medium and fermentation conditions are well-suited for high-density cultivation of B. longum HSBL001 and provide a basis for its industrial application.

The rapid advancement of three-dimensional (3D) printing technologies has significantly expanded their potential applications such as sensors and detector technology. In this study, the gamma-ray shielding performance of ulexite-doped composite resins fabricated via Digital Light Processing (DLP) 3D printing was experimentally investigated to evaluate radiation attenuation capacity. Composite resins containing different ulexite loadings (0, 1, 3, and 5 wt%) were exposed to gamma rays at energies of 356, 662, 1173, and 1333 keV to evaluate their attenuation characteristics.

The small GTPase Rho5 has been shown to be involved in regulating the Baker's yeast response to stress on the cell wall, high medium osmolarity, and reactive oxygen species. These stress conditions trigger a rapid translocation of Rho5 and its dimeric GDP/GTP exchange factor (GEF) to the mitochondrial surface, which was also observed upon glucose starvation. We here show that rho5 deletions affect carbohydrate metabolism both at the transcriptomic and the proteomic level, in addition to cell wall and mitochondrial composition.

The natural tracers δO and δH are essential for tracing hydrological processes by identifying water sources, tracking evaporation loss and floodwater dynamics to enhance water management and flood mitigation strategies. This study employed this approach in the ephemeral, endorheic Cuvelai-Etosha Basin (CEB), spanning northern Namibia and southern Angola, to determine its viability in capturing spatial and temporal hydrological patterns, their timing and interactions during a medium flood condition (2017), and contrasted with a drought year (2014). During the 2017 wet season 219 grab surface water samples were collected from ephemeral waterbodies in four sampling campaigns (February, March, April and May) in addition to a single campaign in May 2014 (63 samples).
