Chronic myelomonocytic leukemia (CMML) is an aggressive hematologic neoplasm characterized by an expansion of CD123 monocytes and plasmacytoid dendritic cells (pDCs). pDC bone marrow clusters in CMML have been associated with higher rates of acute myeloid leukemia transformation. We evaluated tagraxofusp, a CD123-targeted therapy, in a phase 1/2 trial for patients with CMML.

Treatment of patients with platinum-resistant ovarian cancer is a major clinical challenge. We found that high expression of a meiotic protein, Synaptonemal Complex Protein 2 (SYCP2), is associated with platinum resistance and tyrosine kinase ABL1 inhibitor sensitivity in ovarian cancer. We demonstrate that tyrosine kinase ABL1 inhibitors inhibit cancer cell proliferation more efficiently in ovarian cancer cell lines with SYCP2 overexpression.

Epitranscriptomic modifications regulate gene expression and have been implicated in cancer, including breast cancer. Using the SCAN-B cohort, we analyzed 49 messenger RNA modification regulators (mRMPs) across breast cancer subtypes. In the basal subtype, we found significant overexpression of mA readers (IGF2BP1-3), mC regulators (NSUN5, ALYREF, YBX1, YBX2), pseudouridine [PUS1, MARS (or MetRS), RPUSD2], and RNA editing enzymes [APOBEC3A (A3A), A3G, ADAR1], all linked to poor survival.

Introduction: Acute myeloid leukemia (AML) patients are highly susceptible to infection. Moreover, prophylactic and empirical antibiotic treatment during chemotherapy disrupts the gut microbiome, raising the risk for antibiotic-resistant (AR) opportunistic pathogens. There is limited data on risk factors for AR infections or colonization events in treated cancer patients, and no predictive models exist.
