L-3,4-dihydroxyphenylalanine (L-DOPA or Levodopa) is widely used to treat Parkinson's disease, but its large-scale chemical synthesis is costly and environmentally challenging. This study investigates the enzymatic conversion of L-tyrosine to L-Dopa to enhance efficiency and yield without any potential racemization. Tyrosinase, extracted from button mushrooms, was immobilized as cross-linked enzyme aggregates (TY CLEAs), achieving an activity of 31.32 units/g. Commercially available skim milk served as a protein feeder and demonstrated antioxidant properties, helping prevent the formation of L-dopachrome as a byproduct by inhibiting the sequential oxidation of L-dopaquinone, along with ascorbic acid as a reducing agent. We evaluated the stability of TY CLEAs under varying temperatures, pH levels, and organic solvents. High productivity of 1210.95 mgL h was achieved using 10 units of enzyme at a substrate concentration of 15 mM with the use of a surfactant.

As the primary storage protein, highland barley gliadin (HBG) exhibits limitations in the processing of highland barley foods, primarily due to its abundant non-polar amino acids. In this study, HBG was utilized to prepare sugar-HBG complexes with pentose (xylose), hexoses (glucose and galactose), and disaccharides (lactose and maltose) in an aqueous system at a pH of 11 and a temperature of 75 °C. 1,3-Dioleoyl-2-palmitoylglycerol (OPO) is crucial for infant nutrition; however, conventional immobilized lipase requires high-purity enzymes, which increases costs and limits industrial scalability. Herein, Rhizomucor miehei lipase (RML) was immobilized on surface-modified magnetic nanoparticles using cross-linked enzyme aggregates (CLEAs) technology to produce FeO@SiO@TPOAC@RML CLEAs. This approach combines the separation and immobilization of enzymes, allowing for the use of lower-purity lipase, which enhances its suitability for industrial-scale processes.

For purpose of overcoming the negative impact of high-dose phenols on meat quality, xanthan gum (XG), a natural anionic polysaccharide, was employed to prevent the undesirable interaction between myofibrillar protein (MP) and gallic acid (GA, 150 μmol/g) and ameliorate the gel and emulsification characteristics of MP. XG dose-dependently alleviated the structural damage of MP caused by GA and reduced protein aggregation, manifested as the decrease in surface hydrophobicity, turbidity and aggregate size (p < 0.05) and increase in α-helix content and intrinsic fluorescence.

Cross-linked enzyme aggregates (CLEAs) offer a promising strategy for enzyme immobilization, enhancing reusability, high enzyme loading capacity, carrier-free nature, enhanced thermal or operational stability, and ease of preparation and suitability for industrial-scale applications. This study presents the development and optimization of CLEAs of crude amidase from Bacillus smithii IIIMB2907 for efficient biocatalytic synthesis of pharmaceutically important hydroxamic acids from amides. By optimizing key preparation parameters, a robust immobilized biocatalyst was established that retains high activity and stability during repeated use.
