Article Abstract

In this study, a homogeneous immunoassay based on ortho-touch reaction and graphene oxide (GO) quenched acridine esterification chemiluminescence is developed for sensitive detection of SARS-CoV-2 antigen. The sensitivity and specificity of this method was further evaluated with viral protein, virus strains and clinical samples. Benefiting from strong chemiluminescence signal and good quenching performance of GO, this method allows enhanced signal-to-background ratio, and the limit of detection is down to 0.02 ng/ml of viral protein or equivalently 200 copies/ml of virus strains. In clinical evaluations, our approach demonstrated a high level of agreement with digital RT-PCR, achieving a positive coincidence rate of 90.87 %, a negative coincidence rate of 99.77 %, and an overall concordance of 96.53 %, while showing no cross-reactivity with other prevalent viruses. This method offers a promising, sensitive, rapid, and cost-effective platform for early SARS-CoV-2 diagnosis. Moreover, it can potentially be adapted for detecting other pathogens.

The Chinese Society for Therapeutic Radiology Oncology, the Chinese Anti-Cancer Association, the Chinese Society of Clinical Oncology, Head and Neck Cancer International Group, the European Society for Radiotherapy and Oncology, and the American Society for Radiation Oncology jointly developed evidence-based guidelines and a contouring atlas for primary target volume delineation for radiotherapy in nasopharyngeal carcinoma. The guidelines systematically address three crucial challenges: margin design of clinical target volumes; target volume delineation after induction chemotherapy; and low-risk clinical target volume delineation based on local stepwise extension patterns. Based on a comprehensive systematic review and critical appraisal by an international multidisciplinary panel of 50 nasopharyngeal carcinoma specialists from 17 countries and regions, these guidelines are in keeping with advances in nasopharyngeal carcinoma diagnosis and treatment, embodying contemporary treatment concepts, and elaborating on the differences in practice.

The study evaluates the immunomodulatory potential of secukinumab (SECU) and honokiol (HONK) in a murine model of allergic asthma complicated by acute lung injury (ALI), with an emphasis on modulating key inflammatory pathways. The rationale is driven by the necessity to attenuate Th17-mediated cytokine cascades, wherein IL-17 plays a critical role, as well as to explore the adjunctive anti-inflammatory effects of HONK on Th1 cytokine production, including IL-6, TNF-α, and Th2 cytokines. Tick-borne encephalitis virus (TBEV), a highly pathogenic infectious agent that causes serious damage to the nervous system is mainly transmitted by Ixodidae ticks. The laboratory methods (immunoassay and the PCR-based one) are successfully used to detect the virus in tick samples thereby avoiding unwarranted immunoprophylaxis. Although widely used in clinical diagnostics, the sensitivity of electrochemiluminescence (ECL) bead-based immunoassays is intrinsically limited by the reaction mechanism driving the emission of [Ru(bpy)] on the bead surface. Studying the characteristics of T cell activation and cytokine secretion is crucial for understanding the cell-mediated immunological response (CMI). To assess this purpose, we present a droplet-based single-cell immunoassay platform, named Drop-SCIA, which uniquely integrates surface-enhanced Raman spectroscopy (SERS) with homogeneous-phase immunoassay, enabling highly sensitive, multiplexed cytokine detection at the single-cell level and offering superior target enrichment efficiency compared to the widely used interface-based ELISpot assay. Using this platform, we analyzed Jurkat T cell activation and further profiled IL-2 and IFN-γ secretion following coculture with normal breast epithelial cells (MCF-10A) and breast cancer subtypes (MCF-7, MDA-MB-231).
