Type I hypersensitivity reactions are the most common type of hypersensitivity reactions and can cause various symptoms in different body systems. Currently, there is no standard treatment for the different manifestations of type I hypersensitivity reactions at different sites. To address this challenge, a nanocomposite named tFNAs-siRNA-siRNA (TSS) is designed based on tetrahedral framework nucleic acids (tFNAs). It is loaded with two siRNAs and can enter the body through multi-route administration. In terms of the realization of siRNAs' functions, TSS successfully delivers them and downregulates the expression of Gab2 and Syk, thereby inhibiting the migration and release of granules to the extracellular space in RBL-2H3 cells. Interestingly, TSS also exhibits the effect of hindering Ca efflux from mitochondria to stabilize energy metabolism. In the passive cutaneous anaphylaxis and allergic rhinitis model, TSS shows excellent anti-allergic ability, and demonstrates an advantage in intraperitoneal injection, transdermal, and transnasal routes of administration. In sum, TSS offers a new therapeutic strategy for type I hypersensitivity reactions with multiple symptoms, highlighting the application potential of nucleic acids-based multi-route administration delivery system.

Leukocytoclastic vasculitis (LCV) is a type of vasculitis that affects small vessels and is commonly associated with infections, malignancies, drugs, and autoimmune diseases. In this case, a 75-year-old female patient presented with clinical signs of LCV, and after ruling out common etiologies, hydatid disease (HD) emerged as a potential cause. This case highlights the importance of considering parasitic infections, particularly HD, in the differential diagnosis of LCV, especially in regions where these infections are endemic and in patients exposed to relevant environmental risk factors.

Objective: The aim of this study was to evaluate whether fluoroquinolone antibiotics, which are structurally distinct from penicillins, can be safely prescribed as alternatives for patients with a history of immediate-type hypersensitivity reactions (HSRs) to penicillin in the absence of multidrug allergy and without the need for provocation testing.
Methods: We conducted a retrospective analysis of the medical records of patients who presented to the Erciyes University Adult Immunology and Allergy Outpatient Clinic with a documented history of penicillin allergy between 2015 and 2024. Inclusion criteria for immediate hypersensitivity to penicillin included at least one of the following: (1) a history of at least two separate immediate HSRs to the same penicillin; or (2) positive results from penicillin G/V (Penicillin G and Penicillin V) serum-specific immunoglobulin E (SsIgE) and/or skin prick testing.

Objective: The aim of this study was to assess the association between allergic reactions after COVID-19 vaccination and the history of high-risk allergy, individual predisposing factors such as age and gender, and COVID-19 vaccine type.
Materials And Methods: This retrospective cohort study included 234 adult patients (18 years old and above) who underwent a COVID-19 vaccine allergy test up until February 2023 in a Clinic of Allergy and Clinical Immunology in the University Clinical Center of Kosovo. All patients suspected of allergy underwent skin testing: SPT (skin prick test) and IDT (intradermal test) using either an mRNA (ribonucleic messenger acid) vaccine (BNT162b2, Pfizer-BioNTech) and/or an adenoviral vector vaccine (AZD1222, AstraZeneca).
