Cardiac optical mapping provides high-resolution spatiotemporal insights into cardiac electrophysiology. Here, we present a protocol for capturing electrical signals in rats using voltage-sensitive dyes and blebbistatin to inhibit muscle contractions. We describe steps for aortic cannulation, heart isolation, optical mapping, and detailed signal analysis. This staining strategy utilized in this protocol allows for precise signal recording while minimizing motion artifacts, thereby enhancing image quality on the cardiac surface. For complete details on the use and execution of this protocol, please refer to Hu et al..

Purpose: Recent work has shown potential benefits for perimetry with dense spacing. To investigate the impact of normal inhomogeneity of perimetric sensitivity on perimetry with dense spacing, suprathreshold perimetry was used near the optic disc where shadows of blood vessels affect sensitivity in healthy eyes.
Methods: Three groups of participants were tested: 58 healthy older controls, 29 healthy younger controls and 18 patients with glaucoma.

Mentalizing skills-the capacity to attribute mental states-play critical roles in word learning during typical language development. In autism, mentalizing difficulties may constrain word-learning pathways, limiting language-acquisition opportunities. We ask how autistic children encode and retrieve novel words and what drives individual differences.

Background: Tissue clearing techniques combined with light-sheet fluorescence microscopy (LSFM) enable high-resolution 3D imaging of biological structures without physical sectioning. While widely used in neuroscience to determine brain architecture and connectomics, their application for spinal cord mapping remains more limited, posing challenges for studying demyelinating diseases like multiple sclerosis. Myelin visualization in cleared tissues is particularly difficult due to the lipid-removal nature of most clearing protocols, and alternative immunolabeling approaches failed to reach satisfying results.

The human retina exhibits complex cellular heterogeneity which is critical for visual function, yet comprehensive ethnic-specific references are scarce in ophthalmic transcriptomics. The lack of single-cell RNA sequencing (scRNA-seq) data from Asian populations particularly Chinese donors imposes significant limitations in understanding population-specific retinal biology. We constructed the first comprehensive single-cell transcriptomic atlas of the human retina from Chinese donors, generated through high-throughput scRNA-seq of ∼290,000 viable cells obtained from 18 fresh retinal specimens (living donor and post-mortem specimens).

Background: Cardiac issues following radiotherapy are increasingly prevalent among patients with thoracic cancer and coronary disease. However, the mechanisms underlying radiotherapy-induced plaque instability and changes in plaque characteristics on imaging remain unclear. This study used single-cell RNA sequencing to identify key features of vulnerable plaques following radiotherapy.
