Six new prenylated isoflavonoids, named maccosas A-F (1-6), together with ten known prenylated isoflavonoids (7-16) were isolated from the twigs and leaves of Maclura fruticosa. Compounds were isolated by various column chromatographic methods including silica gel, sephadex LH-20, and semi-preparative HPLC. Their structures were elucidated by a combination of 1D and 2D NMR techniques, mass spectrometry, and comparison of their spectral data with those in the literature. Furthermore, compounds 1 and 7 exhibited anti-inflammatory activity with IC values of 4.11 and 1.45 μM. Detection of cellular pyroptosis and western blot assay suggested that compound 1 reduced the number of PI-positive cells and inhibited Nigericin-induced pyroptosis of J774A.1 cells.

Background: Acanthamoeba is a widely distributed genus, causing keratitis, granulomatous amoebic encephalitis and chronic infectious ulcers. At present, there are no effective drugs available for treating Acanthamoeba infections, highlighting the urgent need for the development of novel anti-Acanthamoeba therapies.
Purpose: This study aims to investigate the inhibitory effect of isoliquiritigenin (ISL), a chalcone, and glabridin (GLA), a prenylated isoflavonoid, on Acanthamoeba castellanii trophozoites growth and elucidate their mechanisms of action.

Protein-tyrosine phosphatase 1B (PTP1B) is a master negative regulator of insulin and leptin signaling, yet clinically useful inhibitors remain elusive. Guided by a repurposing strategy, we investigated Tephrosia villosa as a natural source of PTP1B modulators. Chromatographic fractionation of the aerial parts afforded nine flavonoids and related phenolics, whose structures were elucidated by spectroscopic tools.

This is the report of the HPLC-MSn-guided isolation of new anti-inflammatory prenylated isoflavonoids and pterocarpans from the stems of using GNPS molecular networking as the main tool. Cluster analysis guided the isolation of five new prenylated isoflavonoids, diffusiflavone A-E ( and ), two prenylated pterocarpans, diffusicarpan A and B ( and ), and two known compounds 6-prenylorobol () and 3--methylquercetin ().
