Recent advances in fluorescent biosensors have stimulated the development of molecular detection. We herein developed a new orthogonal logic operation of fluorescent biosensor with cell-free to accomplish the detection of atrazine (ATZ) and tetrachlorobiphenyls (PCB77). The transcriptional process to generate fluorescent RNA aptamers (Mango) was controlled by molecules-probe bindings, which regulate split T7 promoter transcription switches ON or OFF. Leveraging the rapid in vitro T7 transcription process and high signal-to-background ratio of the Mango-TO1-Biotin complex, this biosensor demonstrates remarkable sensitivity in detecting ATZ and PCB77, with detection limit of 1.56 pM and 10.2 pM. Moreover, the orthogonality of four logic gates (AND, NOR, INHIBT, NIMPLY) were utilized the ATZ and PCB77 as input to construct, which could be activated by utilizing the target probe-driven association. The output of the fluorescence signal was controlled by split/intact fluorescent RNA aptamer (Mango) to achieve flexible and sensitive orthogonal operations. Significantly, the development of two-input logic gates has enabled the modular detection of various small molecules, offering a promising approach to intelligent multi-input analysis. Predictably, with the advantages of sensitivity, flexibility and easy-to-operate, this orthogonal logic gate platform holds immense potential in small molecules detection.

Static nonreciprocity offers distinct outputs when switching the positions of action and reaction, which is of great interest for designing mechanical logic elements or soft robots. Existing mechanical metamaterials can present specific static nonreciprocal responses, but it remains challenging to obtain multiple and reprogrammable static nonreciprocal modes in a single microstructural topology. Here, a design method of cellular metamaterials is demonstrated via leaving cuts inside metacells, whose contact nonlinearity in the single metacell can offer orthogonal, uniaxial, shear (displacement and Poynting effect) static nonreciprocal modes.

Individuals with mild cognitive impairment (MCI) and an abnormal amyloid biomarker (A+) are at considerable increased risk to develop dementia. Still, these individuals vary greatly in rates of cognitive decline, and the mechanisms underlying this heterogeneity remain largely unclear. The plant synthetic biology toolbox is rapidly expanding; however, there are still limited options for engineering logic gates for the precise modulation of gene expression. CRISPR interference (CRISPRi) represents a promising strategy for engineering logic into plant cells; however, only a limited number of promoter modules have been characterized for CRISPRi-mediated repression. In this study, the transient transgene expression in agroinfiltrated leaves was used to assess the repressibility of a number of promoters with different strengths, including the Figwort Mosaic Virus (FMV) 34S promoter, which showed high repression efficiency using CRISPRi.

The inherent programmability of DNA, dictated by Watson-Crick base-pairing and sequence diversity, renders it a versatile biomolecule for nanoscale engineering. Toehold-mediated strand displacement (TMSD) enhances DNA strand exchange by over 4 orders of magnitude, underpinning applications in molecular circuits, biosensing, and nanodevices. However, its efficiency is constrained by the instability of the toehold duplexes and the reversibility of the initial binding step.
