Because endogenous extracellular vesicles are involved in important physiological functions, various techniques have been developed for their isolation and evaluation. However, methods for evaluating endogenous vesicles within cells are limited. This study presents a technique for the direct extraction and evaluation of intraluminal vesicles (ILVs). This technique combines scanning ion conductance microscopy, electrochemical syringes, and confocal microscopy to extract specific structures within a living cell, achieving high spatial resolution and accuracy at the femtoliter scale. This approach allowed the direct collection of CD63(+) vesicles from HEK293 CD63-pHluorin-RFP cells and showed that their RNA expression profiles were different from those recovered from cytosol and extracellular vesicles isolated by ultracentrifuge. It also identified a subset specifically containing hsa-miR-145-5p and allowed for direct assessment of the local accumulation of miRNAs in cells. This technique is expected to become a powerful tool for evaluating the contents of ILVs within living cells.

This study aimed to histomorphometrically evaluate the effect of guided bone regeneration (GBR) and two implant surfaces on the thickness and height of newly formed bone in dehiscence defects around titanium implants. Three premolars and the first bilateral molar were extracted from ten adult mongrel dogs, and 40 buccal bone dehiscences measuring 5 mm in height and 4 mm in width were created using a University of North Carolina (UNC) periodontal probe to confirm the dimensions. Forty implants were randomly assigned to one of four groups: oxidized implant surfaces (OIS, n = 10), turned/machined implant surfaces (TIS, n = 10), OIS + GBR (n = 10), and TIS + GBR (n = 10).

Unlabelled: There is a need for the development of broad-spectrum antiviral compounds that can act as first-line therapeutic countermeasures to emerging viral infections. Host-directed approaches present a promising avenue of development and carry the benefit of mitigating risks of viral escape mutants. We have previously found the SKI (super killer) complex to be a broad-spectrum, host-target with our lead compound ("UMB18") showing activity against influenza A virus, coronaviruses, and filoviruses.

Background: Parkinson's disease (PD) often presents with lateralized motor symptoms at onset, reflecting asymmetric degeneration of the substantia nigra (SN). Neuromelanin (NM) loss and iron accumulation are hallmarks of SN pathology in PD, but their spatial distribution and interrelationship in PD patients with right-sided (PDR) or left-sided (PDL) motor symptom onset remain unclear.
Purpose: To investigate the spatial vulnerability and interrelationship of NM and iron in the SN among PDR, PDL, and healthy controls (HCs) using MRI.
