Epigenome-wide association studies (EWAS) profile DNA methylation across the human genome to identify associations with diseases and exposures. Most employ Illumina methylation arrays; this platform, however, under-samples interindividual epigenetic variation. The systemic and stable nature of epigenetic variation at correlated regions of systemic interindividual variation (CoRSIVs) should be advantageous to EWAS. Here, we analyze 2,203 published EWAS to determine whether Illumina probes within CoRSIVs are over-represented in the literature. Enrichment of CoRSIV-overlapping probes was observed for most classes of disease, particularly for neurodevelopmental disorders and type 2 diabetes, indicating an opportunity to improve the power of EWAS by over 200- and over 100-fold, respectively. EWAS targeting all known CoRSIVs should accelerate discovery of associations between individual epigenetic variation and risk of disease.

Advanced maternal age increases the risk of pregnancy complications due, in part, to changes in the uterine environment. Here, we show that uterine aging in mice is associated with a progressive increase in transcriptional variation, accompanied by a notable accumulation of activating histone marks at multiple genomic loci. Importantly, the transcriptional signatures of uterine aging differ substantially from senescence markers associated with organismal aging.

The Wuzhishan (WZS) pig, tolerant to high heat and humidity, is an ideal model for genomic study. Summary Answer: Our findings suggest that hCG treatment alters the molecular environment of decidualized ESCs, potentially influencing implantation and immune regulation through epigenetic modifications and changes in the levels of secreted proteins and micro-ribonucleic acids (miRNAs).
What Is Known Already: Embryo implantation depends not only on the quality of the embryo but also on the receptivity of the endometrium, the specialized lining of the uterus that undergoes dynamic changes to support pregnancy. Effective communication between the maternal and fetal compartments, facilitated by molecular signals and cellular interactions, is essential for successful implantation.

Hepatitis C virus (HCV) clearance markedly reduces the risk of hepatocellular carcinoma (HCC); however, HCC continues to develop in a subset of patients, particularly in those with advanced fibrosis or cirrhosis. Leading hepatology societies, including Asian Pacific Association for the Study of the Liver, European Association for the Study of the Liver, American Association for the Study of Liver Diseases, Korean Association for the Study of the Liver, Taiwan Association for the Study of the Liver, and Japan Society of Hepatology, have issued divergent guidelines for HCC surveillance after sustained virologic response, which reflects variations in regional patient populations, healthcare infrastructure, and policy priorities. While traditional risk stratification primarily centers on histological staging of fibrosis, an array of additional host-related factors, including age, sex, alcohol use, metabolic comorbidities, and genetic and epigenetic profiles, further influence individual HCC risks.
