Article Abstract

Ten years after the description of the first cohorts of patients with adenosine deaminase (ADA2) deficiency (DADA2), the pathomechanisms underlying the disease on a cellular level remain poorly understood. With the establishment of the lysosomal localization of the ADA2 protein and its involvement in nucleic acid sensing, the pathophysiologic focus has shifted to the inside of the cell. At the same time, extracellular (serum) ADA2 enzyme activity continues to be the diagnostic reference standard in patients with suspected DADA2. The diverse clinical phenotype and weak genotype-phenotype correlations further complicate the identification of shared cellular mechanisms that cause inflammation, immunodeficiency, and bone marrow failure in the absence of functional ADA2. This review inspects the characteristics of the ADA2 protein and its proposed function. The latter is discussed in the context of possible mechanisms driving the clinical phenotype in patients lacking functional ADA2. We discuss established processes and introduce unexplored pathways in the pathogenesis of DADA2.

Background And Objective: Differentiating central nervous system infections (CNSIs) from brain tumors (BTs) is difficult due to overlapping features and the limited individual indicators, and cerebrospinal fluid (CSF) cytology remains underutilized. To improve differential diagnosis, we developed a model based on 9 early, cost-effective cerebrospinal fluid parameters, including CSF cytology.

Methods: Patients diagnosed with CNSIs or BTs at Xiangya Hospital of Central South University between October 1st, 2017 and March 31st, 2024 were enrolled and divided into the training set and the test set.

Growing evidence have indicated the bidirectional relationships between various inflammatory cytokines and prostate cancer (PCa), but the causality between genetic susceptibility to inflammatory cytokines and PCa was still in initial exploratory phase. This bidirectional Mendelian randomization (MR) research was manipulated to draw causative inferences and the effect of direction between 91 inflammatory cytokines and PCa. Mutations in the human ADAR gene encoding adenosine deaminase acting on RNA 1 (ADAR1) cause Aicardi-Goutières syndrome 6 (AGS6); a severe auto-inflammatory encephalopathy with aberrant interferon (IFN) induction. AdarΔ2-13 null mutant mouse embryos lacking ADAR1 protein die with high levels of IFN-stimulated gene (ISG) transcripts. In Adar Mavs double mutants also lacking the Mitochondrial antiviral signaling (MAVS) adaptor, the aberrant IFN induction is prevented.
