Catalysed C-H activation has emerged as a transformative platform for molecular synthesis and provides new opportunities in drug discovery by late-stage functionalisation (LSF) of complex molecules. Notably, small aliphatic motifs have gained significant interest in medicinal chemistry for their beneficial properties and applications as sp-rich functional group bioisosteres. In this context, we disclose a versatile strategy with broad applicability for the ruthenium-catalysed late-stage meta-C(sp)-H alkylation of pharmaceuticals.

Though N-heterocyclic carbenes (NHCs) have emerged as diverse and powerful discrete functional molecules in pharmaceutics, nanotechnology, and catalysis over decades, the heterogenization of NHCs and their precursors for broader applications in porous materials, like metal-organic frameworks (MOFs), porous coordination polymers (PCPs), covalent-organic frameworks (COFs), porous organic polymers (POPs), and porous organometallic cages (POMCs) was not extensively studied until the last ten years. By or post-synthetic modification (PSM) methods, myriads of NHCs and their precursors containing building blocks were designed and integrated into MOFs, PCPs, COFs, POPs and POMCs to form various structures and porosities. Functionalisation with NHCs and their precursors significantly expands the scope of the potential applications of porous materials by tuning the pore surface chemical/physical properties, providing active sites for binding guest molecules and substrates and realizing recyclability.

Circular tandem repeat proteins ('cTRPs') are de novo designed protein scaffolds (in this and prior studies, based on antiparallel two-helix bundles) that contain repeated protein sequences and structural motifs and form closed circular structures. They can display significant stability and solubility, a wide range of sizes, and are useful as protein display particles for biotechnology applications. However, cTRPs also demonstrate inefficient self-assembly from smaller subunits.
