Article Abstract

Iron plays a prominent role in various biological processes and is an essential element in almost all organisms, including plant-pathogenic fungi. As a transition element, iron occurs in two redox states, Fe and Fe, the transition between which generates distinct reactive oxygen species (ROS) such as HO, OH anions, and toxic OH· radicals. Thus, the redox status of Fe determines ROS formation in pathogen attack and plant defense and governs the outcome of pathogenic interactions. Therefore, spatially resolved visualization of Fe and Fe are essential to understand microbial pathogenesis. Here, we report a simple method for synthesis of the redox-state-selective dyes pyrene-tetramethyl piperidinyl oxyl (p-TEMPO) and 4-(4-methylpiperazine-1)-7-nitrobenz-2-oxa-1,3-diazole (MPNBD) for fluorescence microscopy-based imaging of Fe and Fe ions. Using these dyes, the occurrence and spatial distribution of Fe and Fe ions in vegetative and pathogenic hyphae of the hemibiotrophic maize anthracnose fungus are shown. Vegetatively propagated crops such as cassava, potato, sweetpotato, and yam, or roots and tubers (RTs), play a major role in food security in low- and middle-income countries, yet phytosanitary issues in the tropics lead to substantial yield and quality losses. Challenges to production include institutional limitations that prevent effective responses and potential buildup of pathogens during clonal propagation. Addressing these challenges in a climate change context and diverse sociocultural environments requires a multifaceted approach, including improving access and availability to clean seed by strengthening seed systems; breeding for host resistance and disseminating resistant varieties; strengthening on-farm seed management; and designing effective policies and regulations to deal with seedborne diseases.

Environmental stress adaptation is crucial for the survival and pathogenicity of plant fungal pathogens. In this study, we identified a transcription factor FgMsn2 in Fusarium graminearum, an ortholog of Msn2 in budding yeast. Structural analysis showed that the C2H2 zinc-finger domain is highly conserved across fungi, while other regions are less conserved, suggesting that FgMsn2 may have species-specific functions.
