Article Abstract

T cell bispecific antibodies (TCBs) are a promising new class of therapeutics for relapsed/refractory multiple myeloma. A frequently observed, yet incompletely understood effect of this treatment is the transient reduction of circulating T cell counts, also known as T cell margination (TCM). After administration of the GPRC5D-targeting TCB forimtamig (RG6234), TCM occurred in patients and correlated with cytokine release and soluble B cell maturation antigen decrease. We demonstrate that TCM is accurately represented in the humanized NSG mouse model and occurs at a lower threshold of target expression than systemic cytokine release. Application of whole-mouse tissue clearing and 3D imaging revealed that T cells accumulate in the bone marrow after treatment. We hypothesize that low amounts of targets are sufficient to rapidly redirect T cells upon TCB engagement. Therefore, we propose TCM as a beneficial, highly sensitive and early effect of forimtamig that leads T cells to likely sites of bone marrow tumor lesions.

Cystic teratoma with neuroectodermal cyst and choroid plexus epithelium in the cheek has never been documented in the literature to date. We report the first example of a cystic teratoma with abundant neuroectodermal derivatives, like glial cells and papillary choroid plexus-like epithelium, within the masseteric space and with no intracranial involvement. A two-year-old male child presented with a complaint of a slowly growing swelling on the left side of the face since birth with no history of fever, fatigue or weight loss or other similar swellings elsewhere.

In the absence of effective therapeutics, valve replacement remains the sole intervention for severe calcific aortic valve disease (CAVD), which impacts 26% of adults aged 65 and even 50% of adults aged 85. Pericardium-based bioprosthetic heart valves benefit from marginating anticoagulation. Still, they face recalcification due to residual aldehydes and inadequate elastin stabilization from the glutaraldehyde cross-linking process, posing a significant clinical challenge.

This study presents a multiscale modeling framework for simulating and predicting the behavior and biodistribution of nanoparticles (), focusing on applications such as targeted drug delivery. The framework encompasses two coupled models: (1) a DeepONet-enabled Fokker-Planck equation to model the NP drift-diffusion in the red-blood cell-free layer () that predicts NP margination and concentration profiles taking hematocrit and vessel radius as inputs, built on top of a hemorheological model of shear-induced blood flow and (2) a physiologically based pharmacokinetic (PBPK) model that uses the predicted concentration profiles in microvasculature to inform the biodistribution of NPs across different organ in the body.

Bicavitary epithelioid mesothelioma in a donkey (Equusasinus).

Mesotheliomas are relatively rare and affect many animal species but have not been reported in donkeys (Equus asinus). Here we report a case of bicavitary epithelioid mesothelioma in a 33-year-old female donkey presented with weight loss, lethargy and anorexia. Physical examination revealed tachycardia, ileus and delayed capillary refill time.
