Repeating X-ray computed tomography (CT) measurements over a short period of time allows for obtaining a spatiotemporal four-dimensional (4D) volume image. This study presents an interactive method for segmenting a 4DCT image by fitting a template model to a target organ. The template consists of a three-dimensional (3D) mesh model and free-form-deformation (FFD) cage enclosing the mesh. The user deforms the template by placing multiple curve constraints that specify the boundary shape of the template in 3D space. We also present curve constraints shared over all time frames and interpolated along the time axis to facilitate efficient curve specification. Our method formulates the template deformation using the FFD cage modification, allowing the user to switch between our curve-based method and traditional FFD at any time. To illustrate the feasibility of our method, we show segmentation results in which we could accurately segment three organs from a 4DCT image capturing a swallowing motion. To evaluate the usability of our method, we conducted a user study comparing our curve-based method with the cage-based FFD. We found that the participants finished segmentation in approximately 20% interaction time periods on average with our method.

Non-invasive neuroimaging is important in epilepsy to help identify cerebral abnormalities. Abnormally reduced fractional anisotropy (FA) in deep white matter (WM) from diffusion-weighted imaging (DWI) is widely reported in large multi-cohort studies across all types of epilepsies. However, abnormalities in FA for superficial WM are rarely investigated in epilepsy.

Angelman syndrome (AS) is a debilitating neurodevelopmental disorder caused by loss of maternally-inherited UBE3A. In neurons, paternally-inherited UBE3A is silenced in cis by a long non-coding RNA called Ube3a-ATS. Here, we found that Neisseria meningitidis Cas9 with two mutations (D15A and H587A) in the nuclease domains (dNmCas9) can unsilence the dormant paternal Ube3a allele in mouse and human neurons when targeted to Snord115 snoRNA genes located in introns of Ube3a-ATS.

The concept of micrometer-scale swimming robots, also known as microswimmers, navigating the human body to perform robotic tasks has captured the public imagination and inspired researchers through its numerous representations in popular media. This attention highlights the enormous interest in and potential of this technology for biomedical applications, such as cargo delivery, diagnostics, and minimally invasive surgery, as well as for applications in microfluidics and manufacturing.
