Article Abstract

Background: Rheumatoid arthritis (RA) is an autoimmune disease characterized by synovitis, which can lead to joint deformity. Acupuncture treatment stimulates specific acupoints to adjust qi and blood function, relieving joint inflammation and pain.

Methods: Ultra-high performance liquid chromatography-mass spectrometry (UPLC-QTOF-MS) was utilized for non-targeted metabolomics analysis of plasma samples from the blank group, Adjuvant-Induced Arthritis (AIA) model mice model mice group, and acupuncture group. Metabolite hierarchical clustering analysis, multivariate statistical analysis, standardized processing, principal component analysis (PCA), partial least squares-discriminant analysis (PLS-DA), and other methods were employed to identify targeted metabolites affected by acupuncture treatment in AIA mice. The related metabolic pathways were analyzed using KEGG pathway.

Results: Histopathological results demonstrated that acupuncture at Zusanli point (ST 36) significantly improved the inflammatory response in AIA mice. The PCA score plot indicated relatively close sample clustering within each group with significant differences observed between the four groups, confirming successful establishment of the AIA animal model with metabolic disorders occurring. Acupuncture treatment effectively corrected these metabolic disorders. Plasma metabolomics identified a total of 10 differential metabolites primarily associated with arachidonic acid and pentose phosphate metabolic pathways.

Conclusions: Acupuncture at ST36 can significantly improve the inflammatory response in AIA mice through modulation of arachidonic acid and pentose phosphate metabolic pathways.

The herbal formula Qing-Luo-Yin (QLY) shows potential in activating PPARγ. This study investigated how this property contributed to its anti-rheumatic effects. Some mice with antigen-induced arthritis (AA) were orally treated with QLY and/or PPARγ antagonist/agonist (T0070907/rosiglitazone).

Background: Calcium (Ca) signaling is implicated in the pathogenesis of rheumatoid arthritis (RA). Our previous study found that celastrol (CEL), a natural product derived from herbal plant Tripterygium wilfordii Hook F, suppressed arthritic symptoms in AIA rats via modulating Ca signaling pathway by inhibiting sarcoplasmic/endoplasmic reticulum Ca ATPase (SERCA). However, its high toxicity and narrow therapeutic window severely limit its clinical application.

Rheumatoid arthritis (RA) is a common autoimmune disease characterized by chronic joint inflammation and cartilage damage. The main treatment goal of RA is clinical remission using various kinds of disease-modifying antirheumatic drugs (DMARDs), but there remain unmet medical needs such as the need for sufficient therapeutic efficacy, long-term safety, and affordable treatment. Many preclinical studies suggest that T helper type-17 (Th17) cells have critical roles in the development of RA.

Due to their anti-inflammatory and immunomodulatory capabilities, adipose-derived stem cells (ADSC) are currently considered a promising option for the management of rheumatoid arthritis (RA). To tackle the problems of immunogenicity and tumorigenicity linked to the direct use of cells, current research is focused on the development of effective nanomedicines utilizing ADSC-derived exosomes (ADSC-EXO) for cell-free regenerative medicine. Methotrexate (MTX) was loaded into mesoporous silica through physical adsorption to produce SiO-MTX, with subsequent incorporation into ADSC-EXO via ultrasonication to produce AE@SiO-MTX.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by systemic inflammation and progressive joint destruction. Although current treatments, including conventional disease-modifying antirheumatic drugs (DMARDs) and biologics, offer therapeutic benefits, they are often associated with drug resistance and significant adverse effects with long-term use. Our previous research identified the accumulation of circulating cell-free DNA (cfDNA) as a critical pathogenic factor in RA, contributing to immune dysregulation and sustained inflammatory responses.
