Fibrosis is an abnormal tissue healing process characterized by the excessive accumulation of ECM components, such as COL I and COL III, in response to tissue injury or chronic inflammation. Recent advances in epitranscriptomics have underscored the importance of m6A modification in fibrosis. m6A, the most prevalent modification in eukaryotic RNA, is catalyzed by methyltransferases (e.g., METTL3), removed by demethylases (e.g., FTO), and recognized by reader proteins (e.g., YTHDF1/2). These modifications are crucial in regulating collagen metabolism and associated diseases. Understanding the role of m6A modification in fibrosis and other collagen-related conditions holds promise for developing targeted therapies. This review highlights the latest progress in this area.

Introduction: Lactate has emerged as a multifunctional signaling molecule regulating various physiological and pathological processes. Furthermore, lactylation, a newly identified posttranslational modification triggered by lactate accumulation, plays significant roles in human health and diseases. This study aims to investigate the roles of lactate/lactylation in respiratory diseases.

S-glutathionylation (SSG), a redox-sensitive post-translational modification mediated by glutathione, regulates protein structure and function through reversible disulfide bond formation at cysteine residues. Glutaredoxins (GRXs), pivotal antioxidant enzymes, catalyze SSG dynamics to maintain thiol homeostasis. Recent advances in redox proteomics have revealed that SSG dysregulation is intricately linked to neurodegenerative, cardiovascular, pulmonary and malignant diseases.

Background: This study aimed to investigate the gender-specific associations of skeletal muscle mass and fat mass with non-alcoholic fatty liver disease (NAFLD) and NAFLD-related liver fibrosis in two population-based studies.
Methods: Analyses were based on data from the MEGA (n = 238) and the MEIA study (n = 594) conducted between 2018 and 2023 in Augsburg, Germany. Bioelectrical impedance analysis was used to evaluate relative skeletal muscle mass (rSM) and SM index (SMI) as well as relative fat mass (rFM) and FM index (FMI); furthermore, the fat-to-muscle ratio was built.

Connective tissue growth factor (CTGF) is notably upregulated in scar tissue, making it a promising target for therapeutic intervention. Here, we have designed and screened an antisense oligonucleotide (ASO) that binds specifically to the exon five sequence of CTGF, with particular emphasis on the use of 2'-O-methoxyethyl (MOE) and locked nucleic acid (LNA) modifications to enhance stability and specificity. experiments demonstrated that both MOE-ASO#1 and LNA-ASO#1 significantly inhibited fibroblast proliferation and extracellular matrix protein expression.

Cardiac fibrosis, especially in the infarct border zone, leads to decreased cardiac compliance, impaired systolic and diastolic function, resulting in heart failure. M6A methylation plays a role in fibrosis development. However, its underlying mechanism remains poorly understood.
