(2)- methyl ester and (4)- lactone were recently identified as major components in essential oils and extracts of from Togo. Extended biological evaluation of these acetylenic compounds was however hampered by the reduced amounts isolated. A synthetic route was designed providing access to larger quantities of these two natural products as well as to original non-natural analogs with the prospect of exploring for the first time the structure-activity relationships in this series. Using LC/MS analysis, synthetic samples allowed confirming the presence of the two previously isolated natural products in plant extracts obtained by the accelerated solvent extraction technique. The nematocidal activity of the synthesized compounds confirmed the potency of the natural products, which remain the most active among all analogs tested. The synthesized compounds were also assessed against axenic amastigotes and the HRv pathogenic strain. (2)- methyl ester, (4)- lactone and lactone analogs exhibited the strongest antileishmanial potency. As expected, a longer alkyl chain was necessary to observe significant antimycobacterial activity. The lactone analog bearing a C10 lipophilic appendage displayed the highest antimycobacterial potency. The notable activities of lactones, naturally occurring or analogs, either nematicidal, antileishmanial or antimycobacterial, were compared to their cytotoxicity for mammalian cells and revealed moderate selectivity index values. In this regard, the innocuous (2)- methyl ester and its analogs open up more promising perspectives for the discovery of bioactive agents to protect both agricultural crops and human health.

The study investigated the intrinsic changes in material basis of Angelicae Sinensis Radix during wine processing by headspace-gas chromatography-ion mobility spectrometry(HS-GC-IMS), headspace-solid phase microextraction-gas chromatography-mass spectrometry(HS-SPME-GC-MS), and ultra-high performance liquid chromatography-quadrupole-orbitrap mass spectrometry(UPLC-Q-Orbitrap-MS) combined with chemometrics. HS-GC-IMS fingerprints of Angelicae Sinensis Radix before and after wine processing were established to analyze the variation trends of volatile components and characterize volatile small-molecule substances before and after processing. Principal component analysis(PCA) and orthogonal partial least squares-discriminant analysis(OPLS-DA) were employed for differentiation and difference analysis.

Di(hetero)aroyl dichlorides are desymmetrized upon sequential reaction with alcohols and 2-methyl N-benzyl thiazolium salts within the course of a one-pot three-component reaction yielding ester-substituted aroyl-S,N-ketene acetals under mild conditions in good yields. A prerequisite for the concise one-pot process is the different nucleophilicity of the alcohols and in situ generated S,N-ketene acetals. The resulting compounds are merocyanines with dominant charge-transfer absorption bands which are fluorescent in the solid state, but not in solution.

Chemoselective borylation of methane offers a promising route to organoboron synthesis. We report a porous zirconium UiO-MOF confined mononuclear iron(III) dihydride catalyst enabling efficient monoborylation of CH using HBpin at 185 °C to afford methyl boronic acid pinacol ester in 85% yield with a cumulative TON of up to 921.
