Cilia are antenna-like organelles protruding from the surface of many cell types in the human body. Defects in ciliary structure or function often lead to diseases that are collectively called ciliopathies. Cilia and flagella-associated protein 410 (CFAP410) localizes at the basal body of cilia/flagella and plays essential roles in ciliogenesis, neuronal development and DNA damage repair. It remains unknown how its specific basal body location is achieved. Multiple single amino acid mutations in CFAP410 have been identified in patients with various ciliopathies. One of the mutations, L224P, is located in the C-terminal domain (CTD) of human CFAP410 and causes severe spondylometaphyseal dysplasia, axial (SMDAX). However, the molecular mechanism for how the mutation causes the disorder remains unclear. Here, we report our structural studies on the CTD of CFAP410 from three distantly related organisms, and . The crystal structures reveal that the three proteins all adopt the same conformation as a tetrameric helical bundle. Our work further demonstrates that the tetrameric assembly of the CTD is essential for the correct localization of CFAP410 in , as the L224P mutation that disassembles the tetramer disrupts its basal body localization. Taken together, our studies reveal that the basal body localization of CFAP410 is controlled by the CTD and provide a mechanistic explanation for how the mutation L224P in CFAP410 causes ciliopathies in humans.

Frailty, often linked to sarcopenia, involves reduced muscle strength and mass. While sarcopenia has multiple causes, impaired muscle protein synthesis may contribute. Leucine and resistance training (RT) are anabolic stimuli, but the long-term effects of leucine combined with RT in pre/frail older women remain unclear.

The parasitic protozoan Trypanosoma brucei has a single mitochondrial nucleoid, anchored to the basal body of the flagellum via the tripartite attachment complex (TAC). The detergent-insoluble TAC is essential for mitochondrial genome segregation during cytokinesis. The TAC assembles de novo in a directed way from the probasal body towards the kDNA.

Flaxseed oil contains elevated levels of omega-3 fatty acids (n-3 FA), which have been shown to impact reproductive performance. This study aimed to determine the effects of a flaxseed oil-based supplement on reproductive parameters, feeding behavior, and lipid profile in beef heifers. Sixty Angus and Simmental × Angus heifers (14 months old ± 2 months), blocked by full body weight (BW; 396.

Background: Dry-fermented soybean meal (DFSBM) and wet-fermented soybean meal (WFSBM) were evaluated as alternatives to conventional soybean meal in piglet diets, focusing on growth performance, antioxidant function and fecal microbiome modulation. A total of 225 male piglets (Duroc × (Landrace × Yorkshire)) aged 40 days with a body weight of 13.01 ± 0.

Introduction: This post hoc analysis of an A Toujeo Observational Study (ATOS) aims to evaluate the real-world effectiveness and safety of insulin glargine 300 U/ml (Gla-300) in high-risk subgroups of insulin-naïve people with type 2 diabetes (PwT2D) from multiple geographical regions (Asia, the Middle East, North Africa, Latin America, and Eastern Europe).
Methods: In these post hoc analyses of ATOS, a real-world, 12-month, prospective study included 4422 insulin-naïve adults (age ≥ 18 years) with type 2 diabetes (T2D) uncontrolled (HbA > 7% and ≤ 11%) on one or more oral antidiabetic drugs (OADs) who initiated Gla-300 treatment as per routine practice. Primary and secondary endpoints were studied according to renal impairment (RI) status (without or with) and age group (≥ 70 years).
